摘  要:目的探讨高温致神经管畸形(neural tube defects,NTDs)的分子机制。方法在高温致金黄地鼠NTDs模型的基础上,研究NTDs神经上皮细胞中的表达变化。将孕鼠随机分为实验组和对照组,分别于水浴处理后16、40、64 h(相当于胚龄第8.5、9.5、10.5 d)处死孕鼠,剖腹取鼠胚,应用免疫组织化学染色技术和Western-blot法检测NTDs发生过程中β-catenin在神经上皮细胞中的表达变化。结果免疫组化结果显示:β-catenin在各个时间点实验组和对照组的神经上皮细胞以及周围间充质细胞都有表达,高温处理后16 h和40 hβ-catenin与对照组相比表达减弱(P<0.05)。Western-blot结果显示:高温处理后16和40 h,β-catenin在实验组神经上皮细胞内的表达均比对照组明显减弱;高温处理64 h,实验组与对照组表达无明显差异。结论β-catenin的表达变化与神经管的发育密切相关,其表达降低是高温致神经管畸形发生的重要途径之一。Objective: To explore the molecular mechanism of neural tube defects(NTDs) induced by hyperthermia.Methods: On the basis of the established hyperthermia-induced golden-hamster NTDs model,the expressional changes of β-catenin in the neuroepithelial cells during the occurrence of NTDs were investigated.The pregnant rats were randomly divided into the experimental and control groups.The pregnant rats were sacrificed at16,40 and 64 hours,which were corresponded to the embryonic 8.5,9.5 and 10.5 days of age,and after the water bath treatment,the fetuses were then obtained.The expressions of β-catenin in neuroepithelial cells during the occurrence of NTDs were detected using immunohistochemistry staining technique and Western-blot.Results: The results of immunohistochemistry staining showed that β-catenin in the control and experimental groups was diffusely expressed in the neurepithelium cells and mesenchyme surrounded the neural tube.The expression of hyperthermia-treated groups was significantly decreased at 16 and 40 hours after water bath treatment compared with that of the control group(P〈0.05).The results of Western-blot showed that the expression of β-catenin in the hyperthermia-treated group was significantly decreased at 16 and 40 hours after water bath treatment compared with that of the control group(P〈0.05).Conclushion: The expressional change of β-catenin is closely linked to the development of the neural tube,and the decreased expression ofβ-catenin may be one of the important ways of hyperthermia-induced NTDs.
