摘 要:目的:通过筛选正常气虚、高血压气虚、糖尿病气虚等气虚大鼠垂体高表达且上调的基因,探索气虚证在垂体基因表达层面的物质基础。方法:采用16周龄Wistar大鼠、自发性高血压大鼠(SHR大鼠)、自发性糖尿病大鼠(GK大鼠),综合应用标准化大鼠诊法,及GeneChip Rat Gene 1.0 ST Array等技术,检测正常Wistar大鼠、正常气虚Wistar大鼠、SHR气盛大鼠、SHR气虚大鼠、GK气盛大鼠、GK气虚大鼠等垂体基因的表达,重点关注3个气虚证中垂体基因表达大于1000且较正常一致上调的基因。结果:共筛选得到34个基因:Arhgdia、Arfgap1、Rab 21、Arf5、Arf3、Cdipt、Cds2、Pld3、Ppp3ca、Ppp3cb、Trim35、Itm2b、Itm2c、Tob1、Oaz1、Maged1、Gpc4、Glg1、wdr6、Scarb2、Grn、Hnrpd、Hnrnpab、Snrpb、Gtf2i、Ascl1、Pnrc1、Eef2、Eno2、Gls、Ucp2、Atp6v0c、Pomc、Syp。结论:鉴于以上基因中参与基因转录和翻译有关的基因,以及参与能量代谢相关基因表达活跃,一些调控有关靶器官的功能性基因,如POMC亦表达活跃,且大多与细胞活动有关的基因表达增加,提示气虚证大鼠垂体处于一种积极的代偿状态,这可能是气虚证在垂体层面的物质基础。This study aimed to explore the material base of qi deficiency in pituitary gene expression by screening highly expressed and up-regulated genes in pituitary of qi deficiency rats with hypertension, diabetes mellitus or no disease,. Sixteen weeks old healthy Wistar rats, spontaneously hypertensive rats (SHR rats) and spontaneous diabetic rats (Goto-Kakizaki rats, GK rats) were used. The standardized quantitative four diagnosis methods and GeneChip Rat Gene 1.0 ST Array were adopted to test the pituitary gene expression of normal Wistar rats, normal Wistar rats with qi deficiency, SHR rats with qi deficiency, and GK rats with qi deficiency, especially focusing on the genes with expression more than 1000 and up-regulated consistently in three qi deficiency syndromes compared with normal Wistar rats. Thirty-four genes were obtained, including Arhgdia, Arfgapl, Rab21, Arf5, Arf3, Cdipt, Cds2, Pld3, Ppp3ca, Ppp3cb, Trim35, Itm2b, Itm2c, Tobl ,Oazl, Magedl, Gpc4, Glg1, wdr6, Scarb2, Grn, Hnrpd, Hnrnpab, Snrpb, Gtf2i, Ascll, Pnrcl, Eel2, Eno2, Gls, Ucp2, Atp6v0c, Pomc, Syp. Some up-regulated genes were related to gene transcription and translation as well as energy metabolism. Some functional genes such as POMC were actively expressed; moreover, some genes associated with cell activity were expressed at a higher level. The results suggest that the pituitary is in an actively compensational condition when a rat is suffering from qi deficiency. This may be the material base of qi deficiency in pituitary gene expression.
