摘  要:为比较已构建的鸡传染性法氏囊病病毒(IBDV)重组质粒的免疫原性、进一步研究ChIL-18的分子免疫佐剂作用,本研究将2周龄SPF鸡随机分为6组,以200μg/羽剂量首免:A组免疫重组真核表达重组质粒pCI-VP2/4/3-ChIL-18,B组免疫pCI-ChIL-18-VP2/4/3,C组免疫pCI-co-VP2/4/3-ChIL-18,D组免疫pCI-VP2/4/3,E组免疫pCI-ChIL-18和pCI-VP2/4/3,F组为空白对照,2周后以相同剂量二免,通过观察外周血淋巴细胞增殖效果,测定血清IBDV中和抗体水平、血清ELISA抗体水平和攻毒保护试验等,比较各试验组IBDV DNA疫苗的免疫原性。试验发现:重组质粒pCI-co-VP2/4/3-ChIL-18的保护效果最好,保护率达93.3%;pCI-VP2/4/3-ChIL-18免疫效果次之,保护率达80.0%;pCI-ChIL-18-VP2/4/3与pCI-ChIL-18和pCI-VP2/4/3的免疫效果无显著差异,保护率均为73.3%;但均优于单独使用重组质粒pCI-VP2/4/3组,保护率为60.0%。本实验为进一步研究ChIL-18的分子免疫佐剂作用及研制高效、价廉的IBDV DNA疫苗提供了实验依据。This study evaluated the adjuvant effect of ChIL-18 in several recombinant plasmids,with the aim to develop efficient,inexpensive IBDV DNA vaccines.Two-week-old SPF chickens were randomly divided into 6 groups and immunized with 200 μg recombination plasmids:group A for pCI-VP2/4/3-ChIL-18,group B for pCI-ChIL-18-VP2/4/3,group C for pCI-co-VP2/4/3-ChIL-18,group D for pCI-VP2/4/3,group E for each of pCI-VP2/4/3 and pCI-ChIL-18,group F served as the unvaccinated control.The chickens were boosted with same dosage after two weeks.The immune responses were compared among each testing groups through observing lymphocyte proliferation response of peripheral blood,detecting anti-IBDV neutralization antibody and ELISA antibody level.The results showed that pCI-co-VP2/4/3-ChIL-18 could effectively evoke immune responses in chickens with 93.3% protection rate,followed by pCI-VP2/4/3-ChIL-18 with 80.0% protection rate.There was no significant differences between pCI-ChIL-18-VP2/4/3 and pCI-ChIL-18+pCI-VP2/4/3,both produced 73.3% protection.Immunization with pCI-VP2/4/3 used alone produced 60.0% protection.This study indicated that ChIL-18 could be used as anefficient molecular adjuvant in the development of DNA vaccines against IBDV.
