摘  要:目的:通过构建shRNA沉默HO-1基因,研究抑制HO-1基因表达对胃癌细胞系SGC-7901的影响。方法:构建靶向HO-1基因的shRNA干扰质粒转染人胃癌细胞系SGC-7901,荧光共聚焦显微镜下观察转染效率;应用逆转录聚合酶链反应(RT-PCR)、细胞免疫化学法分别在基因、蛋白质水平检测HO-1基因的抑制效果;应用MTT法检测HO-1基因沉默后胃癌细胞的生长情况。结果:与对照组比较,shRNA在基因、蛋白质水平特异地抑制了胃癌细胞SGC-7901中HO-1的表达,抑制率分别为62.4%、67.6%;HO-1的表达被抑制后,胃癌细胞SGC-7901生长受抑制(P<0.05)。结论:shRNA-HO-1可有效抑制胃癌细胞中HO-1的表达;HO-1的沉默表达可抑制肿瘤细胞生长。Objective: To investigate the effects of inhabiting heme oxygenase-1 on gastric cancer line SGC7901 through heme oxygenase-1 small interference RNA transfection was constructed. Methods: A eukaryotic expression plasmid of shRNA targeting on HO-1 was constructed and was transiently transfected into human gastric cancer line SGC7901.The transfection efficiency was observed under a fluorescence confocal microscopy.Expression of HO-1mRNA and protein were detected by reverse transcription polymerase chain reaction(RT-PCR) and immunocytochemical stainning.Cell proliferation was determined by MTT assay. Results: Compared with control group,HO-1 shRNA effectively inhibited the expression of HO-1mRNAand protein,inhabiting ratio was 62.4% and 67.6% respectively,cell growth was inhabited(P0.05) . Conclusions: HO-1 shRNA effectively inhibited the expression of HO-1 in gastric cancer cells,thus inhibiting the growth of the tumor cells.
