不同药物对人肺巨细胞癌PGCL3细胞表面表达的血小板免疫相关抗原的影响

摘　　要：利用铺有 Ｍａｔｒｉｇｅｌ的 Ｂｏｙｄｅｎ小室观察抗ＣＤ９、抗ＣＤ４２ａ、抗ＣＤ ６３、抗ＴＳＰ抗体对ＰＧ－ＣＬ３细胞侵袭作用的影响；采用流式细胞术观察２．５、１０、２０ｍｇ／Ｌ的川芎嗪、丹参酮ⅡＡ和５０、５００、１０００ Ｕ／Ｉ。的水蛙素和凝血酶对 ＰＧＣＬ３细胞膜表面 ＣＤ９、ＣＤ４２ａ、ＣＤ６３、ＴＳＰ表达的影响，观察川芎嗪、丹参酮ⅡＡ、水蛭素和凝血酶对人肺巨细胞癌ＰＧＣＬ３细胞表面血小板免疫相关抗原ＣＤ９、ＣＤ４２ａ、ＣＤ６３和ＴＳＰ表达的影响，部分阐述各种药物对肿瘤粘附和侵袭影响的不同作用机制。结果抗ＣＤ９抗体明显增加 ＰＧＣＬ３细胞的侵袭，而抗ＣＤ４２ａ、抗ＣＤ６３、抗ＴＳＰ抗体不同程度减少ＰＧＣＬ３细胞的侵袭。川芎嗪可使ＣＤ９＋、ＣＤ４２ａ＋、ＴＳＰ＋细胞的数目减少，ＰＧＣＬ３细胞表面ＣＤ９的表达减少；丹参酮ⅡＡ使ＣＤ９的表达有所增加，ＣＤ６３和 ＣＤ４２ａ的表达有所下降，ＣＤ４２ａ＋、ＴＳＰ＋的细胞数目下降；水蛭素使ＣＤ９、ＣＤ６３和ＣＤ４２ａ的表达有所减弱，ＣＤ４２ａ＋、ＣＤ６３＋细胞的比例下降；凝血酶使ＣＤ９和ＣＤ４２ａ的表达有所减弱，并使ＣＤ４２ａ＋的细胞数目减少。提示川芎嗪、丹参酮ⅡＡ、水蛭?Effects of tetramethylpyrazine (I ), tanshinone Ⅱ A(Ⅱ ), hirudin (Ⅲ ) and thrombin (Ⅳ ) on the expression of CD9, CD24a, CD63 and TSP on human pulmonary giant cell carcinoma (PGCL3) were investigated by Boyden Chamber covered with Matrigel with the aid of flow cytometry. Results showed that anti-CD9 antibody significantly augmented, while the other three antibodies (anti CD42a,anti CD63 and anti TSP), impaired the invasive potentials of PGCL3. I decreased the expression of CD9 and the number of CD9+, CD42a+ and TSP+ cells; Ⅱ increased the expression of CD9, but decreased the expression of CD63 and CD42a as well as the number of CD42a+ and TSP+cells; Ⅲ decreased both the expression of CD9, CD63 and CD42a and the number of CD42a+ and CD63+ cells 4 and Ⅳ decreased the expression of CD9, CD42a and the number of CD42a+ cells. The different effects of these drugs on the expression of antigens immunologically related to platelet on PGCL3 cell surface may be related to their different influential actions on the adhesion and invasion of PGCL3 cells.
