摘 要:目的探讨唑来膦酸刺激人外周血γδT细胞的增殖作用,同时测定增殖后的γδT细胞对人肝癌细胞的杀伤效能。方法以不同浓度的唑来膦酸联合IL-2刺激健康人外周血γδT细胞增殖。以流式细胞仪检测相关表型,3H-TdR掺入法测定细胞增殖程度。以γδT细胞作为效应细胞,分别以不同人肝癌细胞作为靶细胞。以SRB法测定效应细胞对靶细胞的杀伤效能。结果外周血γδT细胞增殖明显,唑来膦酸为1μmol/L浓度时,γδT细胞增殖最明显。所得γδT细胞对3种肝癌细胞均表现出明显的杀伤作用。结论唑来膦酸能明显促进人外周血γδT细胞的增殖,且扩增的γδT细胞在体外对肝癌细胞有显著的杀伤作用。Objective To study the effect of γδT lymphocyte multiplication stimulated by zoledronic acid and to investigate the cytoxicity of γδT lymphocyte to hepatoma carcinoma cell.Methods Different concentration zoledronic acid was exerted to stimulate peripheral blood γδT cells proliferation.Analyze correlated phaenotypes by flow cytometry.The quantity of cells was determined by method of 3H-TdR incorporation.γδT cells were taken as effector cell,and different hepatoma carcinoma cells as target cell.The cytoxicity of effector cells to target cells was determined by method of SRB.Results Peripheral blood γδT cells obviously multiplied,and γδT cells multiplied most obviously at the contraction of 1 μmol/L zoledronic acid.γδT cell displayed powerful cytoxicity to the three kinds of tumour cells.Conclusion Peripheral blood γδT lymphocyte can be stimulated and multiplied by zoledronic acid.The γδT lymphocytes stimulated and multiplied by zoledronic acid have significant cytotoxic activity to hepatoma carcinoma cells in vitro.
