摘  要:目的研究人转铁蛋白(HuTf)与金黄色葡萄球菌肠毒素B(SEB)偶联后,其Tf-SEB偶合蛋白的抗肿瘤活性,探讨Tf作为载体用于肿瘤导向免疫治疗的可能性。方法利用凋亡细胞指数检测和超抗原依赖的细胞介导的细胞毒(SDCC)效应,观察该偶合蛋白的起抗原活性和激活T细胞杀伤肿瘤细胞的作用。结果Tf-SEB偶合蛋白比单体SEB能更有效地激活T细胞的杀伤肿瘤细胞活性,并对MHC-Ⅱ类分子阴性的肿瘤靶细胞也具有较强的杀伤效果。结论Tf作为载体应用于肿瘤的导向治疗具有可行性。Objective Study the anti- tumor activity of human transferrin (HuT f)coupled with superantigen, Staphylococcus aureus enterotoxin B(SEB), and the possibility oftransferrin - targeted in immunotherapy of human tumor. Methods Examined the super-antigen activity of Tf- SEB coupled protein and the effect of Tf- SEB to stimulate T cells tokill tumor cells. By detecting the target cell apoptotic index and the superantigen dependentcell - mediated cytotoxicity (SDCC) to examine the ability of Tf- SEB activating T cells tokill the tumor ce1l. ResuIts By comparing with the SEB, we found that Tf- SEB was moreeffect to activate T cells to kill tumor cells and even kill the MHC - II negative tumor cellsmore significant1y. ConcIusion It is possible for Tf as a carrier to be used in thecarrier - targeted therapy for tumorous diseases.
