摘 要:目的:探讨氧化压力在阿尔茨海默病中是否起着重要作用,以及端粒长度变化及分布是否会因抗氧化治疗而发生改变。方法:将38名阿尔茨海默病(AD)患者平均分为2组(两组患者的年龄、性别、端粒长度、生活水平等因素无明显差异),每组19例,分别给予维生素E与安慰剂。6个月后,测定两组患者尿液中8-异前列腺素F2α(8-iso-PGF2α)的浓度,用甲基化敏感与甲基化不敏感的限制性内切酶HpaII和MspI消化DNA来评估端粒的甲基化程度;分别检测外周血的端粒长度,包括端粒平均长度与端粒长度分布。结果:维生素E治疗组中,患者尿液中8-iso-PGF2α的浓度明显降低;最短端粒的甲基化程度呈现降低趋势。结论:维生素E的抗氧化治疗可以减少AD患者体内氧化压力,最短端粒对氧化压力相对更敏感。Objective:To investigate if the oxidative stress(OS) play a role in the pathogenesis of Alzheimer's disease(AD),if length and distribution of telomeres will change when the patients were administered vitamin E.Methods:Thirty-eight AD patients were selected and divided into two groups(there was no significant difference in age,gender,telomeres length and life level between the two groups).Each group of 19 patients were administered vitamin E and placebo for 6 months respectively.Subtelomeric methylation status was estimated by comparing methylation-sensitive and methylation-insensitive isoschizomer HpaII and MspI related telomere length;telomeres average lengths and percentages of telomere length distribution in peripheral blood mononuclear cells in the two groups were analyzed.Results:The data in vitamin E administration group showed a significant decrease of concentration of urinary 8-iso-PGF2α and a decrease tendency of subtelomeric methylation in shortest telomeres 4.4 kb.whereas no difference in placebo group.Conclusion:The results showed the decrease of OS in the group with vitamin E administration,and the shortest telomeres were more sensitive to OS.
