异氟烷对老年大鼠认知功能及海马NMDA受体NR1亚基磷酸化的影响

摘  要:目的探讨老年大鼠吸入异氟烷后其认知功能变化及NMDA受体NR1亚基磷酸化的分析。方法应用18月龄SD大鼠24只,雌雄各半,随机分成3组:0%异氟烷组(10%)、1.2%异氟烷组(11.2%)和1.8%异氟烷组(11.8%),n=8。对各组大鼠行Morris水迷宫测试,结束用western-blot检测磷酸化NMDA受体NR1亚基。结果11.2%组与10%组在第1天潜伏期比较有显著性差异(P<0.05),第2天以后各时间点潜伏期和空间探索实验的成绩与10%组同时间比较均无差异(P>0.05);11.8%组与10%组潜伏期和空间探索实验的成绩比较均有显著性差异(P<0.05)。11.8%组与10%组NMDA受体磷酸化NR1分析比较有显著性差异(P<0.05)。结论 1.8%异氟烷引起老年大鼠的认知功能下降,推测可能与海马NMDA受体NR1亚基磷酸化降低有关。Objective To study the effects of the different level of isoflurane on the cognitive function and phosphorylation of NMDA receptor 1 subunits on hippocampus in aged rats. Methods Twenty - four eighting - month - old SD rats of either sex were enrolled. They were randomly divided into 3 groups ( n = 8/group ) :the normal control group (10% group) , 11.2% group inhaled 1.2% isofluranne; 11.8% group inhaled 1.8% isofluranne. All rats received isoflurane in a special ass anesthesia box, The Morris water maze testing was perfomled. The left hippocampus was isolated for determination of NMDA receptor 1 subunits expression with Western - blot technique. Results In the place navigation and probe trial performance after 5 days inhaled isoflurane, the latency and frequency decreased significantly in 1.8% isoflurane group compared to that in 0% isoflurane group( P 〈 0.05 ) . In the place navigation and probe trial performance after 5 days inhaled isoflurane, the latency decreased significantly in 1.2% isoflurane group only in the first day compared to that in 0% isoflurane group ( P 〈 0. 05 ). In 1.8 % group , 1.8% isoflnrane decreased the expression of NMDA receptor 1 subunits. Conclusion After inhaled 1.8% isoflurane, there were affected learning and memory performance. NMDA receptor 1 subunits on hippocampus may play a important role in the development of cognitive disturbances.
