QTL analysis of soyasaponin content in soybean

摘  要:利用高皂甙含量大豆材料和低皂甙含量大豆材料为亲本组配F2群体,以SSR分子标记技术对F2代群体与皂甙含量相关的QTL进行定位,以便为选育高皂甙含量的特种大豆品种提供理论依据。利用皂甙含量高的哈91016与皂甙含量低的N98-9445A大豆杂交获得的F2代群体,用500对SSR引物对大豆皂甙含量进行QTL定位,其中多态性标记有106个。QTL分析结果表明,控制大豆皂甙含量的QTL分别位于连锁群MLG K、MLG D1a上的Sat_044和Satt580附近,其LOD值分别为2.09和2.87,Sat_044与Satt102的遗传距离是11.4 cM,Satt580与Sat_036的遗传距离是18.7 cM;其对皂甙含量的贡献率分别为12.6%和15.8%。The quantitative analysis method of soybean soyasaponins was studied, which used F2 populations whose parents were of the high soyasaponin content and the low soyasaponin content soybean materials by SSR markers to locate genes about saponin content, which provided a theoretical basis for the high soyasaponin content of soybean varieties breeding. In the study, F2 progeny segregated from a cross between HA-91016 and N98-9445A were used to screen 500 SSR markers. A total of 106 SSR markers which were polymorphic and clearly segregated were used for subsequent QTLs analysis. QTL The result showed that QTLs which controlled the content of soybean saponins were mapped on the linkage groups in MLG K, MLG Dla, and LOD of Sat_044 and Satt580, were 2.09 and 2.87, respectively, the distance between Satt102 and Sat_044 was 11.4 cM, and the distance between Satt580 and Sat_036 was 18.7 cM, which accounted for 12.6% and 15.8% of the phenotypic variation in soyasaponin content in this population.
