动物源大肠杆菌染色体ampC基因突变与头孢菌素耐药关系研究

摘  要:为检测动物源性大肠杆菌染色体ampC基因突变与头孢菌素类药物耐药性的关系,选取22株头孢菌素类药物耐药菌作为受试菌株,采用K-B纸片法测定了其对7种头孢菌素的敏感性,采用PCR测序的方法进行受试菌染色体ampC基因调控区和部分编码区碱基突变分析.结果表明:受试菌对7种头孢菌素的耐药率均在50%以上,20株受试菌ampC基因调控区和部分编码区碱基发生了突变,占90.9%;共有10种突变组合,突变频率最高的为+185,+58,+122,+126,-88,-82,-18,-1位.ampC基因调控区和部分编码区碱基突变与β-内酰胺类药物耐药性有关.22 cephalosporin-resistive Escherichia coli were selected to detect the relationship between the mutation of the control and mero-coding region of the chromosome ampC gene and the resistance of the Cephalosporin.The sensitivity to 7 Cephalosporin were determined by Kirby-Bauer,the control and mero-coding region of the chromosome ampC gene were amplified by PCR and sequenced.The resistance of Cephalosporin were above 50%.20 bacteria(90.9%)existed mutation and had 10 mutation combination,the mutation commonly appeared in +185,+58,+122,+126,-88,-82,-18,-1.The resistance to cephalosporin was related to the mutation in the chromosome ampC control and mero-coding region.
