辛伐他汀对糖尿病大鼠视网膜ICAM-1表达的影响

摘　　要：目的探讨糖尿病大鼠视网膜血管细胞间粘附分子-1(ICAM-1)的表达变化,观察辛伐他汀(simvastatin)对视网膜屏障(BRB)破坏的治疗作用。方法 SD大鼠30只,随机分为正常对照组(CON组),糖尿病组(DM组),辛伐他汀治疗组(SIM)。尾静脉注射链脲佐菌素(streptozocin,STZ)建立糖尿病模型,3个月后处死动物。用伊凡思蓝方法检测视网膜的渗透性;用酶联免疫吸附实验检测ICAM-1的表达。结果造模后3个月SIM组和DM组大鼠视网膜血管渗透性较CON组显著增加(P<0.01),组间比较有差异(P<0.01)DM组大鼠视网膜ICAM-1蛋白水平较CON组相比增加了1.15倍(P<0.01);SIM组糖尿病大鼠视网膜ICAM-1蛋白水平均较DM组减少了29%(P<0.01)。结论视网膜ICAM-1的表达增强参与了BRB的破坏,辛伐他汀可减轻BRB破坏。Objective Discuss the changing expression of ICAM-1 in retina of Streptozocin（STZ） induced diabetes mellitus（DM）,to observe the effect of sinvastatin on the expression of ICAM-1 and the non-lipid therapy of sinvastatin in the pathogenesis of diabetic retinopathy.Methods Thirty adult SD rats were randomly divided into normal control group（CON）,sinvastatin treatment（SIM）,and DM normal sodium（DM） groups.Diabetic model was established by injection through vena caudalis with streptozotocin.Simvastatin（20mg/kg·d） was administered by gavages from the second day in 10 rats and the equivalent amount of normal saline solution was used at the same way in other 10 model rats.Vascular permeability was quantified by analyzing albumin leakage using Evans blue method,and the protein levels of ICAM-1 in the retinas were detected by Elisa.Results After three months,the leakage of EB in retina was increased in diabetic groups compared with that in the control group,peritoneal injection with sinvastatin decreased the leakage of EB in retina.After peritoneal injection of simvastatin,the protein levels of ICAM-1 were increased by 15% in DM group.The protein levels o ICAM-1 were decreased by 29% in SIM group to DM group.Conclusions sinvastatin can protect the blood-retinal barrier of diabetic rats.It can also decrease the protein levels of ICAM-1.
