摘 要:目的研究浒苔水溶性多糖的分离提取、组成及生物活性。方法经水煮、醇析、真空干燥得到浒苔粗多糖,由苯酚-硫酸比色法测定其中水溶性多糖含量;用气相色谱-质谱法和红外光谱对其化学成分进行分析。以抗坏血酸作为对照品,同时采用生物化学法测定了浒苔多糖对羟基自由基和超氧阴离子等自由基的清除能力。结果浒苔粗多糖提取率为10.25%,其中水溶性多糖含量为86.1%。浒苔水溶性多糖为硫酸多糖,对羟基自由基和超氧阴离子具有良好的清除能力,且大小与多糖的用量呈正相相关性,且比抗坏血酸的活性强。结论浒苔多糖为硫酸杂多糖,具有显著的清除自由基的能力。Objective To study the isolation,purification,composition and biological activity of water-soluble polysaccharide from enteromorpha prolifera. Methods Crude polysaccharide from enteromorpha prolifera was obtained by extracting with hot water and precipitating with ethanol. Water-soluble polysaccharides content was determined by phenol-vitriolic colorimetry. The chemical compositions of the product were examined by the gas chromatography-mass spectrometric and infrared spectrum. Meanwhile,the scavenging effects of the prepared polysaccharides on hydroxyl radical and superoxide radical were measured by biochemical method with ascorbic acid as a control. Results The extraction percentage of crude polysaccharide was 10. 25% with 86. 1% of water-soluble polysaccharide. The main components of the cude polysaccharide were sulfated polysaccharides. The scavenging effects of sulfated polysaccharides on ·OH and O2-· were significant and concentration dependent,better than ascorbic acid. Conclusion Polysaccharide from enteromorpha prolifera is sulfated heteropolysaccharide,and its biological activity is significant.
