摘 要:2-甲基-6-植基-1,4-苯醌甲基转移酶(MPBQ-MT,VTE3)是维生素E合成途径中的关键酶之一,通过转化模式植物拟南芥,研究大豆VTE3(GmVTE3)基因在拟南芥中过量表达对维生素E组分含量的影响。成功构建了植物表达载体pZP212-GmVTE3,将其转入模式植物拟南芥,获得转基因纯系植株6个;利用HPLC检测技术对转基因拟南芥种子和叶片中维生素E含量进行了测定。结果表明:6个转基因纯系植株种子中,γ-生育酚/δ-生育酚比值均比野生型有所提高,为野生型的1.7倍;叶片中未检测到δ-生育酚。表明GmVTE3可以有效地把δ-生育酚转化为γ-生育酚,从而改变植物体内VE的组成。MPBQ-MT (2-methyl-6-phytyl-1,4-benzoquinone methyltransferase,VTE3 ) was a key enzyme in the biosynthesis of vitamin E,GmVTE 3 was transformed intoArabidopsis thaliana for investigating the effect of its overexpression on the content and composition of vitamin E. In the study,plant expression vector pZP212-GmVTE3 was constructed forArabidopsis transformation,and obtained six homozygous lines,and vitamin E were measured in the seeds and leaves of transgenic lines by HPLC. The results showed that the ratio of γ -tocopherol / δ-tocopherol in the seeds of six transgenic homozygous lines was 1.7 times of wild type,and in the leaves δ-tocopherol was undetectable,thus indicating thatGmVTE 3 gene was able to increase the content of γ -tocopherol and decrease the content of δ-tocopherol,and then changed the content and composition of vitamin E.
