摘 要:目的观察复方血栓通对人视网膜色素上皮(RPE)细胞(CRL-2302)增殖、移行以及表达血管内皮生长因子(VEGF)和基质金属蛋白酶2(MMP-2)的影响,探讨其作用途径及机制。方法复方血栓通作用于CRL-2302细胞,采用MTS比色法,观察细胞增殖的情况;Transwell小室检测细胞移行的影响;Western blot和实时荧光定量RT-PCR方法分别检测细胞VEGF、MMP-2蛋白和mRNA的表达。结果 MTS比色法显示,一定浓度的复方血栓通胶囊(1.562 5~200 g/L)对VEGF诱导的CRL-2302细胞增殖具有抑制作用;Transwell小室实验显示,复方血栓通胶囊浓度为0,3.125,6.25和12.5 g/L时CRL-2302细胞移行数分别为183.67±13.32,145.67±14.57,87.33±29.14和34.67±7.51;3.125,6.25和12.5 g/L浓度的复方血栓通胶囊使CRL-2302细胞VEGF、MMP-2蛋白和mRNA的表达均显著降低。结论复方血栓通胶囊能够抑制RPE细胞的增殖、移行,这可能是其抑制RPE细胞参与血管生成的途径;其机制与抑制VEGF和MMP-2表达密切相关。Objective It is to observe the influence of Complex Xueshuantong on proliferation,migration,and expression of vascular endothelial growth factor(VEGF) and matrix metalloproteinase 2(MMP-2) of human retinal pigment epithelial(RPE) cell line(CRL-2302) cells in vitro,in order to understand the action pathway and mechanism.Methods MTS method was used to access the effect of Complex Xueshuantong on the proliferation of CRL-2302 cells induced by VEGF.Transwell insert was used to detect the migration of CRL-2302 cells.Western Blot and real-time quantitative reverse transcription polymerase chain reaction(RT-PCR) were used to detect the expression of VEGF,matrix metalloproteinase 2(MMP-2) proteins and mRNA of CRL-2302 cells.Results The proliferation of CRL-2302 cells induced by VEGF was inhibited by Complex Xueshuantong at concentrations of from 1.562 5 to 200 g/L.When the concentration of Complex Xueshuantong was 0,3.125,6.25,and 12.5 g/L,cells number that migrated to under the Transwell membrane was 183.67±13.32,145.67±14.57,87.33±29.14,and 34.67±7.51 respectively.At concentrations of 3.125,6.25,and 12.5 g/L,Complex Xueshuantong could inhibit the expression of VEGF and MMP-2 proteins and mRNA of CRL-2302 cells.Conclusion Complex Xueshuantong could inhibit CNV concerned with CRL-2302 cells through the path of suppression of proliferation,migration of the cells.The inhibition of the expression of VEGF and MMP-2 proteins and mRNA of CRL-2302 cells may be one of the important mechanisms.
