毕赤酵母密码子用法对人骨形成蛋白6表达的影响分析

摘　　要：对已筛选出的毕赤酵母阳性转化子菌株(His+Mut+)和(His+Muts)进行PCR验证,用SDS-PAGE和Western blot方法检测重组骨形成蛋白6的表达情况,为了解释无表达的结果,对酵母的密码子与重组骨形成蛋白6的密码子兼容性进行了理论上的初步分析。The screened strains （His＋Mut＋）and（His＋Muts）haboring multi-copy of human bone morphogenetic protein-6 expression cassette is confirmed by PCR.SDS-PAGE silver staining and western blot are applied to detect expression of recombinant human bone morphogenetic protein-6.Since there is no indication of expression,a theoretical analysis is performed to study the compatibility of gene code between Pichia Pastoris and human bone morphogenetic protein-6.The results show that the synonymous codon of glutamic acid and arginine of Pichia Pastoris is different from that in human bone morphogenetic protein-6.
