摘 要:茉莉酸(JA)和水杨酸(SA)是植物中重要的内源激素和防御反应信号分子,快速而准确地定量测定植物内源SA和JA含量非常重要。以番茄叶片作为材料,研究利用气相色谱(GC)同时测定植物内源JA和SA的含量。利用丙酮/柠檬酸(50mmol/L)(体积分数=7/3)和乙酸乙酯提取番茄叶片中的JA和SA,提取液经N2吹干后用三甲基硅烷化重氮甲烷甲酯化,然后用顶空-固相微萃取(HS-SPME)方法进行收集,正己烷洗脱后进行气相色谱分析,用氢离子火焰检测器(FID)进行检测,梯度升温程序为60℃(1min)至250℃,15℃/min,250℃,3min。从样品中得到与茉莉酸甲酯(MeJA)和水杨酸甲酯(MeSA)标准品完全吻合的色谱峰。该方法可以广泛用于同时测定植物内源的SA和JA含量。Jasmonic and salicylic acids are important phytohormones and plant defensive signaling compounds.Here a new method was developed for the simultaneous quantification of the two signaling compounds in tomato leaves by using headspace-solid phase microextraction(HS-SPME) and gas chromatography with hydrogen ion flame detector(FID).Leaf samples were extracted by mixture of acetone and citric acid(50 mmol/L)(V/V=7/3),and ethyl acetate.Then the supernatant was dried by N2 and subsequently methylated with trimethylsilyldiazomethane.The volatilized compounds were collected by using headspace-solid phase microextraction on Tenax adsorbents,and eluted with n-hexane.Eluted samples were analyzed by using GC with hydrogen ion flame detector(FID).The temperature gradient was increased from 60 ℃(1 min) to 250 ℃ in a rate of 15 ℃/min and held on 3 min at 250 ℃.The final chromatographic peaks of JA and SA in the samples were identical to the authentic compounds.This method will be useful for simultaneous quantification of JA and SA in plants.
