摘 要:[目的]探索口腔扁平苔藓的中医证候研究方法,探寻阴虚火旺型口腔扁平苔藓的miRNAs表达特征。[方法]选取3例阴虚火旺型口腔扁平苔藓患者及3例正常人血液,分离提取miRNAs,荧光标记后与miRNAs基因芯片杂交,SAM软件筛选阴虚火旺型口腔扁平苔藓患者和正常人有表达差异的miRNA。[结果]获得5个口腔扁平苔藓特异表达miRNAs,其中3个下调,2个上调。[结论]基因芯片技术可用于口腔扁平苔藓中医证候研究,hsa-miR-18a上调和hsa-miR-99b下调可能是阴虚火旺型口腔扁平苔藓的标志性miRNAs。[Objective] To observe the method of researching TCM syndrome of OLP,and to identify the specific miRNAs associated with OLP of hyperactivity fire due to yin deficiency type.[Methods] The miRNAs were extracted and isolated from six samples of blood,including 3 cases with OLP of hyperactivity fire due to yin deficiency type and 3 cases of the control group.miRNAs microarrays were used for screening the expression differences of miRNAs.Data were analyzed using SAM software.[Results] A total of 5 differentially expressed miRNAs were found to be associated with OLP,3 miRNAs were downregulated,2 miRNAs were up-regulated.[Conclusion] Gene chip could be used for the research on TCM syndrome of OLP,miR-18a and miR-99b may be act as the key regulators of gene expression in OLP and correlated with pathogenesis of OLP.
