摘  要:目的纯化芡实多糖并测定其含量。方法采用Sevag法和三氯乙酸法纯化芡实多糖,并以硫酸-蒽酮法显色,紫外分光光度法测定其含量。结果以三氯乙酸法脱蛋白的多糖损失率为25.75%,以Sevag法脱蛋白的多糖损失率为56.91%;吸收度与葡萄糖浓度在2.0-18.0μg/ml范围内呈良好线性关系,线性回归方程为A=0.0414 C-0.0331(r=0.999),平均加样回收率为100.2%,RSD=0.4%(n=6)。纯化后粗多糖中多糖含量为73.27%。结论三氯乙酸法用于芡实粗多糖的脱蛋白是可行的。硫酸-蒽酮法简便、准确、重现性好,可用于芡实中多糖的含量测定。Objective To purify and determine the contents of polysaccharide in Semen Eurylie.Methods The polysaccharide was purified by sevag method and trichloroacetic acid method.The contents of polysaccharide were determined by sulfuric acid-anthrone method and detected at 500 nm by UV spectrophotometer.Results The loss ratio of polysaccharide with the TCA method was 25.75%,and with the Sevag method was 56.91%.A good linear relationship was achieved over the concentration range of 2.0~18.0μg/ml for glucose with an average recovery of 100.2%,RSD=0.4%(n=6).The equation of linear regression was A=0.0414C-0.0331(r=0.999).The content of polysaccharide in the crude purified polysaccharide was 73.27%.Conclusion Trichloroacetic acid method is reasonable.Sulfuric acid-anthrone method is accurate and reproducible,and it can be used to determine the contents of polysaccharide in Semen Eurylie.
