GeM_(10)体外对PHA诱发的肿瘤病人外周血单个核细胞产生IL-2的影响

摘　　要：ＧｅＭ１０是一种抗癌生物碱－金属配位药物，动物实验发现ＧｅＭ１０对实验肿瘤Ｓ１８０肉瘤有抑制作用，能明显延长艾氏腹水癌荷瘤小鼠的存活时间，封闭荷瘤鼠体单核巨噬细胞系统，ＧｅＭ１０的抑癌作用解除。观察了ＧｅＭ１０对ＰＨＡ诱发的肿瘤患者外周血单个核细胞（ＰＢＭＣ）产生白介素－２（ＩＬ－２）能力的影响。结果发现ＧｅＭ１０能提高肿瘤患者外周血单个核细胞产生白细胞介素－２的能力（对照组：１．７９２±０．８２３；ＧｅＭ１０６０μｇ／ｍｌ２．９６５±１．１８３；Ｐ＜０．０５）。提示：ＧｅＭ１０的抗癌作用与其刺激机体免疫活性细胞。GeM 10 is a new organic germanium compound-alkalaid-metal complex.Our previous date showed that GeM 10 could inhibited S 180 Solid tumor and Ehrilish ascites in mice.Although GeM 10 showed strong antitumor effect in Vivo,the mechanism of the antitumor effect is still unclear.in this paper,We studied the cytokine(IL 2) production by the peripheral blood mononuclear cells(PBMC) by Il 2 dependent cell line CTLL 2 and MTT colorometric method.It is observed that GeM 10 could enhanced the IL 2 production of peripheral blood mononuclear cells of 12 tumor patients (control:1.792+0.823 to 60μg/ml GeM 10 2.965=1.183 P<0 05).these finding suggest that GeM 10 can inhibit the tumor growth and may have promise as a antitumor drug and biological response modifier in humans
