摘  要:观察8-氯腺苷对人白血病细胞HL-60和人T淋巴母细胞白血病细胞株(Molt-4)的分化和凋亡作用以及两种诱导效应的关系。流式细胞术-DNA(FCM-DNA)测定显示8-氯腺苷使HL-60和人T淋巴母细胞白血病细胞株Molt-4阻滞在G0/G1期。8-氯腺苷诱导人急性粒白血病细胞HL-60分化,表现为硝基蓝四氮唑(nitrobluetetrazolium,NBT)阳性细胞增多及出现形态学分化表型。通过形态学观察,FCM-DNA测定及琼脂糖电泳分析结果表明,8-氯腺苷可以诱导Molt-4细胞凋亡,且有时间、剂量依赖性,但未发现8-氯腺苷对HL-60细胞有诱导凋亡的作用。8-Chloroadenosine (8-Cl-Ado) is a new compound that is able to inhibit the growth of a wide variety of cancer cell types in vitro and in vivo. In this study we have found that differentiation of human acute myeloblastic leukemia(M2)cell line HL-60 can be induced by 8-Cl-Ado, FCM-DNA analysis showed that 8-Cl-Ado caused a block in G0/G1 phase of HL-60 and Molt-4 lymphoblast cell line. 8-Cl-Ado induced dose and time-dependent apoptosis in Molt-4 cells , but apoptosis was not observed in HL-60 cells treated with 8-Cl-Ado. These results suggest that induction of differentiation and apoptosis by 8-Cl-Ado is cell type specificity,and there is no causality between differentiation and apoptosis.
