FAP-1与子宫内膜癌抵抗Fas介导的细胞凋亡的关系

摘　　要：目的探讨Fas相关磷酸酯酶-1(FAP-1)在子宫内膜癌抵抗Fas介导的细胞凋亡中的作用。方法应用S-P免疫组织化学方法检测63例子宫内膜癌中FAP-1、Fas及Fas-L的表达,同时检测13例子宫内膜非典型增生及20例正常子宫内膜组织作为对照。结果 FAP-1在子宫内膜癌中表达率为71.4%,明显高于正常子宫内膜中的表达率25.0%(P<0.05)。Fas在子宫内膜癌和正常子宫内膜组织中表达率分别为52.4%和85.0%(P<0.05)。Fas-L在子宫内膜癌和正常子宫内膜组织中表达率分别为76.2%和35.0%(P<0.05)。Fas蛋白表达阳性、阴性者子宫内膜癌组织中,FAP-1表达阳性率分别为51.5%和93.3%,两者间呈明显的负相关(P<0.05);而Fas-L表达阳性、阴性者中,FAP-1表达率分别为81.3%和40.0%,两者之间呈明显的正相关(P<0.05)。结论子宫内膜癌抵抗Fas介导的细胞凋亡与肿瘤细胞表达FAP-1有关,FAP-1引起的凋亡抑制在子宫内膜癌的发生、发展中起一定作用。Objective To study the FAP-1 expression and its relationship with resistance against fas-mediated apoptosis in endometrial carcinoma. MethodsUsing streptavidin-biotin peroxidase （S-P） immunohistochemical method,we examined the expression of FAP-1,Fas and Fas-L proteins in 63 endometrial carcinoma tissues, 13 endometrial atypical hyperplasia tissues and 20 normal endometrial tissues. ResultsPositive FAP-1 expression was observed in 45/63 （71.4% ） cases of endometrial carcinomas, whose positive rate was higher than normal endometrial epithelium （25%, P 0.05）. It was the same as Fas-L expression （76.2%versus 35%, P 0.05）.The positive rate of Fas in endometrial carcinoma was lower than normal endometrial epithelium （52.4%versus 85%, P 0.05）. The positive rate of FAP-1 was higher in Fas-negative than Fas-positive carcinoma cases （93.3%versus 51.5% , P 0.05）.There was significant difference in FAP-1 expression between Fas-L positive and negative carcinoma tissues（81.3% versus 40%, P 0.05）.ConclusionsThe expression of FAP-1 was significantly correlated with resistance against Fas-mediated apoptosis in endometrial carcinoma.The inhibitory role of FAP-1 in apoptosis contributed to the endometrial carcinogenesis and subsequent progression.
