摘 要:目的:研究角叉菜不同提取物的抑菌和免疫活性。方法:将角叉菜的乙醇提取物依次用石油醚、乙酸乙酯、正丁醇进行萃取,取石油醚相进行硅胶柱层析得到9种提取物P1-P9,对9种提取物进行抑菌(大肠杆菌、金黄色葡萄球菌、面包酵母菌和假丝酵母菌)活性的研究,通过小鼠脾淋巴细胞增殖实验和NO释放实验检测免疫活性。结果:抑菌实验结果表明,P4、P6、P7、P8均能抑制大肠杆菌和金黄色葡萄球菌的生长,P3和P5只能抑制金黄色葡萄球菌的生长,所有的提取物均不能抑制面包酵母菌和假丝酵母菌,P5、P6、P7、P8对大肠杆菌的最低抑菌浓度(MIC)分别为25、50、25、12.5 mg/mL,P3、P4、P5、P6、P7、P8对金黄色葡萄球菌的MIC分别为6.25、12.5、25、100、50、6.25 mg/mL;免疫实验表明,9种角叉菜提取物对小鼠免疫细胞具有不同的刺激能力,有些提取物能显著促进脾淋巴细胞的增殖,并能促进小鼠腹腔巨噬释放NO。结论:角叉菜提取物有一定的抑菌和免疫活性。Objective: To study the antimircrobial and immunomodulation actitvities of extracts from Chondrus ocellatus.Methods: The samples were extracted with ethanol,and then extracted with petroleum ether,ethyl acetate,n-butanol.Nine kinds of extracts(P1 to P9) were prepared by silica gel column chromatography from extraction of petroleum ether of Chondrus ocellatus.Effect on growth of Escherichia coli,Staphylococcus aureus,yeast bread and candida were studied.T-lymphocytes and macrophage cells were obtained from healthy mice for improved lymphopoiesis test as well as NO production of murine peritioneal macrophages.Results: P4,P6,P7,P8 could inhibit the growth of Staphylococcus aureus and Escherichia coli,but P3 and P5 could only inhibit the growth of Staphylococcus aureus.All extracts had no effect on yeast bread and candida.The minimum inhibitory concentration(MIC) of P5,P6,P7,P8 to Escherichia coli were 25,50,25,12.5 mg/mL,and the MIC of P3,P4,P5,P6,P7,P8 to Staphylococcus aureus were 6.25,12.5,25,100,50,6.25 mg/mL.Nine exacts had stimulating activities on cells mediated immunity in mice.Some extracts could significantly promote spleen T-lymphocyte proliferation and NO production of murine peritioneal macrophages.Con-clusion: The conclusion is that some extracts have antimircrobial antivitie and can advance the immunity of healthy mice.
