养血清脑颗粒对大鼠肝CYP 1A2活性的影响

摘　　要：目的:研究养血清脑颗粒对大鼠肝CYP1A2活性的影响。方法:实验分为空白对照、酶诱导、酶抑制及养血清脑组,以非那西丁作为探针药物。大鼠分别给予生理盐水、酶诱导剂、酶抑制剂和养血清脑颗粒,灌胃7天后,静脉注射非那西丁,不同时间点采集血样,用高效液相色谱法测定血浆中非那西丁的浓度,用Topfit 2.0软件估算其药动学参数。结果:空白对照、酶诱导、酶抑制和养血清脑组探针药物非那西丁的t1/2分别为(0.44±0.05)、(0.27±0.02)、(0.87±0.34)、(0.41±0.04)h;CL分别为(21.78±3.57)、(31.04±2.83)、(12.77±3.26)、(18.32±1.61)mL·min-1。结论:养血清脑颗粒对大鼠肝CYP1A2无明显的抑制或诱导作用。OBJECTIVE：To investigate the effects of Yangxue qingnao granule on the activity of hepatic CYP 1A2 in rats.METHODS：These rats were divided into blank control group,enzyme induction group,enzyme inhibition group and Yangxue qingnao group.Phenacetin as a probe drug was given to all rats by intravenous injection.4 groups were given medicine respectively for 7 days.The blood samples were collected at different time.HPLC method was used to determine the concentrations of phenacetin in rat plasma,and pharmacokinetic parameters were estimated by Topfit 2.0 software.RESULTS：The t 1/2 values of phenacetin in 4 groups were（0.44±0.05）h,（0.27±0.02）h,（0.87±0.34）h,（0.41±0.04）h,respectively.And the total clearance values of it were（21.78±3.57）mL·min -1,（31.04±2.83）mL·min -1,（12.77±3.26）mL·min -1,（18.32±1.61）mL·min -1,respectively.CONCLUSION：Yangxue qingnao granule has no significant effects on the activity of hepatic CYP 1A2 in rats.
