摘  要:目的探讨选择性COX-2抑制剂Celebrex体外对人卵巢癌细胞株HO8910增殖及体内对裸鼠卵巢癌原位移植瘤模型生长的抑制效应。方法采用四甲基偶氮唑盐(MTT)法检测Celebrex对人卵巢癌细胞株HO8910生长的影响。建立人卵巢癌原位移植瘤模型,给予Celebrex8周,采用免疫组织化学法和RT-PCR法检测移植瘤组织中COX-2的表达。结果 (1)MTT检测结果显示,COX-2抑制剂与HO8910细胞增殖之间有剂量-效应和时间-效应依赖性;(2)Celebrex干预可抑制裸鼠卵巢癌原位移植瘤生长;(3)免疫组织化学染色和RT-PCR检测结果表明,Ce-lebrex干预组COX-2表达下调(P<0.05)。结论 COX-2在裸鼠卵巢癌原位移植瘤中表达,COX-2抑制剂在体内体外对卵巢癌细胞生长均有抑制作用。Objective To study the selective COX-2 inhibitor( Celebrex ) on growth of ovarian cancer cells and evaluate the effects of the Celebrex on the transplanted ovarian cancer of the nude mice. Methods The inhibitory effects of Celebrex on the growth of HO-8910 were studied in vitro by MTT method. Meanwhile,the inhibition of tumor growth in transplanted ovarian cancer of the nude mice treated by Celebrex. The expression of COX-2 in transplanted ovarian cancer tissue of the nude mice was determined by immunohisochemisty and RT-PCR. Results ( 1) MTT study showed that Celebrex produced a dose-dependent inhibition and a time-dependent inhibition of HO-8910 proliferation. ( 2) The growth of transplanted ovarian cancer was down-regulated by he treatment of Celebrex. ( 3) The expression of COX2 determined by immunohisochemisty and RT-PCR in transplanted ovarian cancer tissue was strongly down-regulated by the treatment of Celebrex ( P 0. 05) . Conclusion Conclusions COX-2 expresses in the transplanted ovarian cancer tissue of the nude mice. Selective COX-2 inhibitor( Celebrex ) can significantly inhibit ovarian cancer in vivo and in vitro.
