摘 要:目的探讨河北省汉族人群D17S30位点的基因频率分布,为法医的基因认定提供依据。方法用聚合酶链式反应(PCR)对河北汉族人群201名无关个体D17S30位点进行扩增,扩增产物采用聚丙烯酰胺凝胶电泳及溴化乙锭染色,依据扩增片段长度确定各等位基因,分别使用POMS软件和自编软件,进行统计学分析和遗传多态性分析,并对10个家系资料进行了分析。结果共检出12种等位基因,45种基因型,基因型分布符合Hardy-Weinberg平衡定律(χ2=10.6872,P=0.7104,df=14)。观测杂合度ho为75.12%,DP、EPP和PIC值分别为0.9611、0.7280和0.8493。该位点呈共显性遗传。结论D17S30位点在河北汉族人群中的基因频率分布具有良好的多态性,适用于该人群的个人识别和亲子鉴定。ObjectiveTo explore D17S30 locus distribution in Han population of Hebei province and provide clues of gene discrimination for forensic analysis.MethodsPolymerase chain reaction (PCR) was performed followed by polyacrylamide gel electrophoresis and ethidium bromide staining to determine each allele according to the length. Then,POMS software and another software programmed by ourselves were used separately to make the χ 2 test and genetic polymorphism analysis. Besides, 10 families were analyzed. Results45 phenotypes and 12 alleles ranging from 168-938 bp were observed among the 201 unrelated individuals. Alleles 4, 1 and 5 were the most common alleles in Hebei Han population. No deviation from Hardy-Weinberg equilibrium was found (χ 2= 10.687 2 , P = 0.710 4 , df=14). The observed heterozygosity (h o), discrimination power (DP), excluding probability of paternity (EPP) and polymorphism information content (PIC) were 75.12 %, 0.961 1 , 0.728 0 and 0.849 3 respectively. Family analyses showed this locus was in good agreement with codominant inheritance. ConclusionThe better polymorphism of the D17S30 locus makes it suitable for individual identification and paternity diagnosis in Han population of Hebei province.
