孕酮、他莫西芬对子宫内膜癌细胞增殖及雌激素受体相关受体表达的影响

摘　　要：目的探讨孕酮及他莫西芬（tamoxifen,TAM）对子宫内膜癌细胞生长的影响及其对雌激素受体相关受体（ERRα）表达的影响。方法体外培养子宫内膜癌Ishikawa细胞,采用MTT法检测不同浓度孕酮及TAM对子宫内膜癌细胞生长的影响,同时采用Western Blot方法检测细胞中ERRα蛋白水平的变化。结果不同浓度孕酮对Ishikawa细胞体外增殖具有抑制作用,呈剂量-时间依赖性。TAM对Ishikawa细胞体外增殖具有双向作用,在×10-9~×10-7mol/L TAM作用下促进细胞增殖,并呈剂量-时间依赖性;在×10-5mol/L TAM作用下抑制细胞增殖。×10-9~10-6mol/L TAM作用后下调细胞中ERRα蛋白表达,×10-5mol/L TAM及孕酮作用后上调ERRα蛋白表达。结论孕酮及高浓度他莫西芬（×10-5mol/L）可抑制子宫内膜癌细胞的增殖,这种抑制作用可能通过调控细胞中ERRα表达实现;而低浓度他莫西芬则促进子宫内膜癌细胞的增殖。Objective To explore the effect of progesterone and tamoxifen on cell proliferation and the expression of estrogen receptor related receptor （ERRα） of endometrial carcinoma. Methods Endometrial carcinoma cell line Ishikawa cells were cultured in vitro. MTT assay was performed to measure the cell proliferation. The levels of ERRα protein were measured by Western Blot. Results Different concentrations of progesterone inhibited the cell proliferation of Ishikawa cells in a dose and time-dependent manner.The lower concentrations of tamoxifen （×10-9~×10-7mol/L） stimulated cell proliferation in a dose and time-dependent pattern, while the higher concentration of tamoxifen （×10-5mol/L） inhibited cell proliferation. The lower concentrations of tamoxifen （×10-9~×10-7mol/L） might down-regulate the protein expression of ERRα and the higher concentration of tamoxifen （×10 -5mol/L） and different concentrations of progesterone might up-regulate the protein expression of ERRα. Conclusions Progesterone and the higher concentration of tamoxifen may inhibit the cell proliferation of endometrial carcinoma by regulating the expression of ERRα. The lower concentrations of tamoxifen may stimulate cell proliferation of endometrial carcinoma.
