摘  要:目的 探讨类固醇生成因子-1(SF-1)在原发性醛固酮增多症(PA)发病中的作用.方法 分别收集16例醛固酮瘤组织标本和12例正常肾上腺组织标本,Western b1ot法检测各标本SF-1蛋白表达水平,荧光定量聚合酶链反应(PCR)法检测其mRNA表达水平.结果 醛固酮瘤组SF-1无论在蛋白表达水平还是在mRNA表达水平均高于正常肾上腺组,蛋白表达水平分别为(0.87±0.05和0.39±0.07,P<0.01),mRNA水平则前者是后者的10.48倍(P<0.01),差异均有统计学意义.结论 SF-1表达上调在PA的发病中发挥着重要作用.Objective To explore the effect of steroidogenic factor-1 ( SF-1 ) on the pathogenesis of primary aldosteronism (PA). Methods Sixteen aldosteronoma and twelve normal adrenal gland specimens were collected. Western blotting and real-time polymerase chain reaction (PCR) were used to detect the protein and mRNA expression of two groups, respectively. Results No matter in protein or mRNA level, the contents of SF-1 in aldosteronoma group were higher than that in normal adrenal gland group. Protein expression were 0. 87 ± 0. 05 and 0. 39 ± 0. 07 ( P 〈 0. 01 ). In mRNA expression level, the former was 10. 48 times higher than that of the latter ( P 〈 0. 01 ). There was significant difference between the two groups. Conclusion The up-regulation of SF-1 playsan important role in PA, and it may be become as a molecular target to study the pathogenesis of PA.
