摘  要:目的观察大鼠反复癫痫发作后心室肌β受体mRNA表达水平的动态变化。方法将20只健康SD大鼠随机分为4组,对照组、1周组、2周组和4周组。对照组大鼠只在头部植入电极,不给电刺激,其余各组用电休克诱发癫痫发作。在相应时间点处死大鼠,取左心室组织,用RT-PCR方法测定β1、β2受体mRNA的表达水平。结果大鼠反复癫痫发作后β1受体mRNA的表达水平明显下降(P<0.05),β2受体mRNA的表达水平无明显变化。β2受体在β受体中所占比例由对照组的15%上升到癫痫反复诱发4周后的40%。结论大鼠反复癫痫发作后心脏β2受体在β受体中所占比例逐渐升高。Objective To investigate the effects of repeated seizures on the expression of cardiac beta-adrenoreceptor(AR) mRNA levels.Methods Twenty Sprague-Dawley rats were randomly divided into four groups: control group,1-,2-and 4-week groups.They were subjected daily to maximal electroshock(MES) for a total of 1 week to 4 weeks.Ventricular beta 1-AR and beta 2-AR mRNA levels were measured by reverse transcription-polymerase chain reaction(RT-PCR).Results In comparison with control group,beta 1-AR mRNA levels decreased gradually(P0.05),whereas the change of beta 2-AR mRNA levels was not significant.The ratio between beta 2-AR and beta AR increased from 15% to 40%(P0.01).Conclusion The ratio between beta 2-AR and beta-AR increases gradually after repeated seizures.
