摘  要:目的观察苦参碱对体外培养的人ACHN肾癌细胞的生长抑制作用,并探讨其可能的作用机制。方法应用不同浓度的苦参碱分别作用于人肾癌ACHN细胞株24、48、72、96 h,采用MTT法观察苦参碱对ACHN的细胞毒作用;透射电镜和流式细胞术检测苦参碱对ACHN细胞凋亡的影响;免疫细胞化学技术检测苦参碱对ACHN细胞Bcl-2、Bax蛋白表达的影响。结果不同浓度的苦参碱对ACHN细胞株均有一定的增殖抑制作用,且呈量效和时效关系;苦参碱能明显诱导ACHN细胞株凋亡;苦参碱作用后,ACHN细胞株Bax蛋白表达明显增强,Bcl-2蛋白表达有下降趋势,但无统计学差异。结论苦参碱对体外培养的人肾癌ACHN细胞株有显著的增殖抑制作用,其机制可能与调节Bcl-2/Bax蛋白表达比值、诱导细胞凋亡有关。Objective To observe matrine's inhibitory effect on the growth of ACHN human renal cell carcinoma in vitro and explore its possible mechanism.Methods The cell line of ACHN human renal cell carcinoma was treated with matrine of different concentrations for 24,48,72 and 96 h,respectively.MTT assay was used to evaluate the cytotoxic effects of matrine on ACHN cells.The ACHN cell apoptosis induced by matrine was detected with transmission electron microscopy and flow cytometry.The expressions of Bcl-2 and Bax proteins were evaluated with immunocytochemical technique.Results Matrine of different concentrations had a cytotoxic effect on ACHN cell line in obvious dose-and time-dependent manners.Matrine could obviously induce apoptosis of ACHN cell line.Bax protein expression was significantly increased while the expression of Bcl-2 protein declined but with no significant difference.Conclusion Matrine has an obvious inhibitory effect on proliferation of ACHN renal cell carcinoma in vitro,which may be related to down-regulating the ratio of Bcl-2/ Bax protein expression and promoting cell apoptosis.
