摘 要:目的研究奥曲肽对bFGF诱导的兔晶状体上皮细胞增殖的影响。方法体外培养的兔晶状体上皮细胞分为对照组和实验组,其中对照组包括空白对照和bFGF对照组,实验组包括单独用奥曲肽和联合10mg·mL-1碱性成纤维细胞生长因子(basic fibroblastgrowth factor,bFGF)组,奥曲肽浓度分别为10-11mol.L-1、10-10mol.L-1、10-9mol.L-1、10-8mol.L-1。加药后48h用MTT比色法测定奥曲肽对细胞增殖的抑制作用;用流式细胞仪分析细胞周期。结果 bFGF组与空白对照组相比,吸光度A值升高,加入奥曲肽作用48h后,随着药物浓度增加,A值明显降低(P<0.05),抑制率增加,10-9mol.L-1、10-8mol.L-1奥曲肽组的抑制率分别为(24.7±3.6)%、(22.8±6.6)%。联合应用bFGF+奥曲肽组与仅用bFGF组相比,吸光度下降更明显(P<0.05),即抑制率增加更明显,10-9mol.L-1、10-8mol.L-1奥曲肽联合bFGF组的抑制率分别为(32.1±2.9)%、(33.6±3.3)%。流式细胞仪分析奥曲肽对兔晶状体上皮细胞的周期的影响与MTT法栓测结果呈现大致相同趋势,随着药物浓度的增加,处于静止期的细胞逐渐增加而增殖期细胞逐渐减少。结论奥曲肽可明显抑制bFGF诱导的兔晶状体上皮细胞的增殖,并呈剂量依赖性关系,为临床筛选防治后发性白内障的药物提供一定的依据。Objective To investigate the effects of octreotide on lens epithelial cells(LEC)proliferation induced by bFGF in rabbit.Methods Rabbit's LEC cultured in vitro were divided into control group and experimental group,then the control group were subdivided into blank control group and bFGF-induced control group,the experimental group were subdivided into single octreotide group and combined bFGF(10 mg·L-1)group,the concentrations of octreotide were 10-11 mol·L-1,10-10 mol·L-1,10-9 mol·L-1,10-8 mol·L-1.The inhibition of octreotide on rabbit's LEC proliferation was detected by MTT method at 48 hours after octreotide was added and the cell cycle was analyzed with flow cytometry.Results Compared with blank control group,the absorbance(A)value was elevated in bFGF group,the A values were obviously declined with the increase of octreotide concentration at 48 hours after octreotide was added(P0.05),the inhibitive rates were increased,which of 10-9 mol·L-1,10-8 mol·L-1 octreotide were(24.7±3.6)%,(22.8±6.6)%.Compared with single octreotide group,the A values were obviously decreased(P0.05)and the inhibitive rates were significant increased,which of 110-9 mol·L-1,10-8 mol·L-1 octreotide were(32.1±2.9)%,(33.6±3.3)%.The inhibitive effects of octreotide on LEC detected by with flow cytometry were similar to that detected by MTT,the stationary LEC were gradually increased and the proliferative LEC were gradually decreased with the increase of octreotide concentration.Conclusion Octreotide can effectively inhibit the LEC proliferation induced by bFGF at a dose-dependent manner,which offers certain basis for choosing the medicine to prevent and treat the posterior capsular opacification.
