摘　　要：目的:探讨RNA干扰沉默EZH2基因对人卵巢癌细胞系A2780增殖和迁移的影响。方法:真核表达载体介导靶向沉默EZH2 shRNA的重组质粒转染A2780细胞,Real-time RT-PCR检测EZH2基因的沉默效果,MTT法检测细胞增殖,流式细胞仪检测细胞周期,Transwell小室检测细胞迁移。结果:EZH2shRNA转染明显下调EZH2的表达(P<0.001)。重组质粒稳定转染至A2780细胞后,细胞增殖速度明显下降(P<0.05),G0/G1期细胞增加(P<0.01),体外迁移能力下降(P<0.01),差异均有统计学意义。结论:EZH2基因沉默能有效抑制卵巢癌细胞的增殖和迁移能力,为卵巢癌的表观遗传学治疗提供新靶点。Objective：To investigate the effect of EZH2 knockdown on cell proliferation and migration in ovarian cancer cell line A2780 by RNA interference.Methods：After the shRNA plasmid vector targeting EZH2 was transferred into A2780 cells,the mRNA expression of EZH2 was determined by Real-time RT-PCR,cell proliferation and migration capability were examined by MTT assays and Transwell migration assay,cell cycle was analyzed by Flow Cytometry.Results：EZH2 shRNA transference down regulated EZH2 mRNA successfully（P〈0.001）.EZH2 knockdown caused G0/G1 cell cycle arrest,suppressed cell proliferation capability as well as cell migration capability of A2780 cells（P〈0.05）.Conclusion：RNA interference silencing EZH2 gene can significantly inhibit cell proliferation and migration of A2780 cells,thus provide a novel target for epigenetic therapy of ovarian cancer.
