摘　　要：研究旨在利用生物反应器原理获得大量的、供实验研究的人瘦素蛋白。根据GenBank公布的人瘦素蛋白基因序列(登录号为NM000230)设计一对特异性引物,构建鸡输卵管瘦素特异性表达载体pOV3-OB,将重组质粒采用PEI包裹后翅静脉注射试验用鸡,收集蛋清并检测瘦素蛋白表达情况。结果显示:通过Western-blot检测,瘦素蛋白发生特异性表达。人瘦素蛋白可以在鸡输卵管中实现暂态表达,本研究为该蛋白的获取及其生物活性的研究奠定基础。This paper aimed at obtaining a large number of available human leptin protein for experiment research in the principle of bio-reactor. According to the published sequence of human leptin gene in GenBank （accession number is NM000230）,we desiged a pair of specific primer to construct the chicken oviduct-specific expression of leptin vector POV3-OB. The recombinant plasmid was injected into the wing vein of tested chickens after being wrapped with PEI. Egg was collected and detected to exam whether the leptin protein is expressed. The gene expression product was analyzed by Western-blot,results showed that leptin proteins expressed specificly. This experiment showed that the recombinant plasmid could express human leptin protein in chicken oviduct. It made the basis tot the obtaining of the large amount of protein and research of its biological activity.
