摘 要:以苯乙烯(St)为功能单体,二乙烯基苯(DVB)为交联剂,采用固液联合致孔的方式,制备了疏水多孔载体。将多孔载体用于脂肪酶Candida sp.99-125的固定化,得780U/g的固定化活力。固定化使酶在30~50℃范围内热稳定性得到了明显的提高,最适反应温度提高了5℃,在pH6.0~9.0范围内酶的相对活力有所提高,最适pH不变仍为8.0。固定化酶单次催化油酸和十六醇的酯化率达98%,其操作稳定性好,连续间歇操作15批,酯化率仍可达到50%。A macroporous hydrophobic poly(styrene-divinylbenzene) support has been prepared using a multiple porogen and used for the immobilization of Candida sp. 99-125 lipase. The conditions of the lipase immobilization on the support were optimized and an optimum activity (780 U/g) was obtained. After immobilization,the thermal stability of the lipase was increased in the temperature range 30-50 ℃,and its stability in the pH range 6.0-9.0 was also increased. The optimum temperature after immobilization was increased by 5 ℃. The enzyme preparation had increased relative activity at pH 6.0-9.0 and a maximum activity at pH 8.0. The esterification reaction of cetyl alcohol and oleic acid was efficiently catalyzed by the enzyme preparation with the yield of ester being 98%. Operational stability tests indicated that the ester yield was still as high as 50% after 15 batch cycles.
