嗜麦芽寡养单胞菌发酵产聚羟基烷酸酯的研究

摘  要:嗜麦芽寡养单胞菌(Stenotrophomonas maltophlia)HG-B-1是一株可产PHAs的菌株。文章采用逐因子试验和响应面分析法优化S.maltophlia HG-B-1的发酵工艺条件。首先考察了菌株HG-B-1的生长曲线,得到其生长周期约为24h。在此基础上,运用逐因子试验确定其生长的最适碳源和氮源,分别为麦芽糖和NH4Cl;然后在单因素试验的基础上采用三因素五水平的响应面分析,根据回归分析确定较优的发酵工艺条件为:温度35.4℃,pH7.9,摇床转速129rmin-1。采用该工艺条件发酵24h,得到OD600为2.966,是理论值的98.8%。Stenotrophomonas maltophlia HG-B-1 was a strain for polyhydroxyalkanoates producing.Fermentation condition of S.maltophlia HG-B-1 was optimized by seriatim-factorial experiment,response surface methodology and monofactorial experiment.The optimum carbon and nitrogen for HG-B-1 were obtained by seriatim-factorial experiment,which were maltose and NH4Cl respectively.Experiment factors and levels were firstly selected by one-factor test,then central-composite design and response surface analysis with three factors and five levels were used,and factors affecting the culture condition were determined by regression analysis.The results showed that the optimum condition were:35.4 ℃,pH 7.9,129 r?min-1.After 24 h incubation under the optimized condition,a maximum OD600 2.966 was obtained (98.8 % of theory).
