摘 要:目的研究下调核因子-κB(NF-κB)/p65的表达对舌鳞状细胞癌Tca8113细胞的增殖、周期及迁移能力的影响,并探讨其相关的分子机制。方法应用脂质体2000将NF-κB/p65 siRNA转染舌鳞状细胞癌Tca8113细胞,用Real-ti me PCR和Westernblotting技术分别检测p65 mRNA及其蛋白的表达,采用CCK-8试剂盒和流式细胞术分别检测NF-κB/p65 siRNA对Tca8113细胞增殖和细胞周期的影响,Boyden小室法分析其对Tca8113细胞迁移能力的影响。进一步用Real-ti me PCR和Western blotting技术检测细胞周期相关基因cyclin E和cdk2以及浸润转移相关因子MMP-9 mRNA和蛋白表达的变化。结果 NF-κB/p65 siRNA能有效下调舌鳞状细胞癌Tca8113细胞中NF-κB/p65 mRNA和蛋白的表达(P<0.05),明显抑制Tca8113的细胞增殖(P<0.05),诱导细胞静止在G0/G1期,并降低Tca8113细胞的迁移能力。NF-κB/p65转染Tca8113后的48h,cyclin E、cdk2及MMP-9的表达均显著降低(P<0.05)。结论 NF-κB/p65在舌鳞状细胞癌细胞周期及浸润转移中起重要作用,这可能与相关基因表达的改变有关。Objective To study the effects of down-regulation of nuclear factor--κB (NF--κB)/p65 expression on cell proliferation, cell cycle and cell migration, and explore the possible molecular mechanism in Tca8113 cells, a tongue squamous cell carcinoma celt line. Methods NF--κB/p65 siRNA was transfected into tongue squamous cell carcinoma cell line Tca8113 cells using lipofectamine 2000, and the expressions of p65 mRNA and protein were detected by Real-time PCR and Western blotting after transfection with NF--κB/p65 siRNA. Effects of NF--κB/p65 siRNA on cell proliferation, cell cycle and celi migration of Tea8113 ceils were investigated with CCK-8 kit, flow cytometry and Boyden chamber, respectively. Furthermore, the changes in expressions of cell cycle related genes cyclin E and cdk2 and invasiveness and metastasis related gene MMP-9 were analyzed with Real-time PCR and Western blotting methods. Results Introduction of NF--κB/p65 siRNA into TcaSll3 cells resulted in obvious down regulation of expressions of NF--κB/p65 mRNA and protein (P〈0. 05), and obviously inhibited cell proliferation of Tca8113 cells (P〈0.05), arrested cell cycle at G0/G1 phase, and decreased the ability of cell migration of Tca8113. Moreover, the expressions of cyclin E, cdk2 and MMP9 declined significantly 48 hours after transfection of Tca8113 with NF -κB/p65 siRNA. Conclusions NF-κB/p65 plays an essential role in the regulation of cell cycle, invasiveness and metastasis in tongue squamous cell carcinoma, which may be associated closely with the changes in related genes expressions. It is expected to provide a theoretical basis for molecular target therapy of tongue squamous cell carcinoma in clinical setting.
