白细胞介素-18基因转染的NCI-H460肺癌细胞生物学特性研究

摘　　要：目的:研究IL-18基因转染的NCI-H460肺癌细胞的生物学特性。方法:设转染组(GT组)、空载体转染组(PT组)和未转染组(NT组),分别以IL-18转染的NCI-H460细胞、pcDNA3.1(+)转染的NCI-H460细胞及未转染的NCI-H460细胞为实验细胞,观察3组细胞生长情况;MTT法测定细胞增殖。Boy-den Chamber试验比较细胞侵袭能力。将3种细胞接种裸鼠,观察成瘤时间、生存时间及肿瘤大小。结果:培养第7天3组的细胞数量为GT组(5.92±0.27)×104,PT组(7.20±0.20)×104,NT组(7.09±0.12)×104,72h时A值为GT组0.97±0.08,PT组1.35±0.07,NT组1.28±0.10,3组穿透滤膜细胞数为GT组27.50±5.72,PT组38.67±6.31,NT组41.33±5.89,GT组细胞增殖及侵袭力低于PT组及NT组。3组成瘤时间为GT组(12.0±2.19)d、PT组(9.17±1.94)d、NT组(8.67±1.86)d,生存时间为GT组(51.67±7.45)d、PT组(40.17±9.83)d及NT组(37.83±10.07)d,肿瘤体积为GT组(2082.4±670.2)mm3、PT组(3893.4±316.3)mm3、NT组(3338.2±725.4)mm3,GT组细胞在裸鼠体内生长速度低于PT组及NT组。结论:IL-18基因转染的NCI-H460肺癌细胞增殖减慢,侵袭性减弱,致瘤性下降,为IL-18用于治疗恶性肿瘤奠定基础。OBJECTIVE：To investigate the biological property of interleukin-18 gene transfected by NCI-H460 lung cancer cells.METHODS：Three groups including GT,PV and NT were set.NCI-H460 cells transfected byIL-18,pcDNA3.1（＋） and non-transfected NCI-H460 cells were assigned for the 3 group respectively.The cell growth of the 3 group was observed.Cell proliferation was assayed with MTT method.Boyden Chamber test was used to determine cell invasive capacity.BALB/c mice were inoculated with cancer cells mentioned above.The time of tumor-forming,mice survival and tumor size were observed.RESULTS：On the 7th day of culture,the amounts of cells were（5.92±0.27）×104 for Group GT,（7.20±0.20）×104 for Group PT and （7.09±0.12）×104 for Group NT.The A values in 72 h were 0.97±0.08 for Group GT,1.35±0.07 for Group PT and 1.28±0.10 for Group NT.The cells penetrated filter membrane were 27.50±5.72 for Group GT,38.67±6.31 for Group PT and 41.33±5.89 for Group NT.The tumor-forming periods were （12.0±2.19） d for Group GT,（9.17±1.94） d for Group PT and （8.67±1.86） d for Group NT.The survival time was （51.67±7.45） d for Group GT,（40.17±9.83）d for Group PT and （37.83±10.07）d for Group NT.The tumor sizes of Group GT,group PT and group NT were（2 082.4±670.2）mm3,（3 893.4±316.3） mm3 and （3 338.2±725.4） mm3,respectively.The tumor growth velocity of Group GT was lower than those of Group PT and Group NT.CONCLUSION：IL-18 gene transfected NCI-H460 cells grow and proliferate slowly with weakened invasive capacity and oncogenicity.
