胃癌组织Survivin和PTTG表达相关性及其临床意义的研究

摘　　要：目的:探讨胃癌组织中生存素(Survivin)及垂体瘤转化基因(PTTG)的表达相关性及其临床意义。方法:采用RT-PCR检测48例人胃癌组织及相应的癌旁组织中Survivin的表达情况;采用免疫组织化学(SP)染色法检测48例人胃癌组织及相应的癌旁组织中PTTG的表达情况。分析Survivin和PTTG表达与胃癌临床病理因素的关系及两者的相关性。结果:Survivin在胃癌组织中的阳性表达率60.4%(29/48)明显高于其在癌旁组织中的表达率22.9%(11/48),χ2=13.886,P<0.05。Survivin的表达与胃癌组织的TNM分期及有无淋巴结转移密切相关(P<0.05)。PTTG在胃癌组织中的阳性表达率为54.2%(26/48)明显高于其癌旁组织中的阳性表达率12.5%(6/48),z=-3.890,P<0.05。PTTG的表达与胃癌组织的分化程度、TNM分期及有无淋巴结转移密切相关(P<0.05)。研究显示,Survivin与PTTG呈正相关(rs=0.555,P<0.01)。结论:胃癌组织中Survivin与PTTG的表达均上调,在胃癌的发生发展中起重要作用,两者可能存在共同的激活机制,构成肿瘤细胞凋亡的多种途径。两者的检测可作为反映胃癌生物学行为和判断预后的有效指标之一。OBJECTIVE：To investigate the expressions of Survivin,PTTG and their clinical significance in gastric cancer tissues.METHODS：Expressions of Survivin in 48 gastric cancer groups and adjacent nontumorous gastric group were detected by RT-PCR,Meanwhile,expressions of PTTG in 48 gastric cancer group and adjacent nontumorous gastric group were detected by Streptavidin-Peroxidase immunohistochemistry staining.Analysis of Survivin and PTTG expression and clinicopathological factors of gastric cancer relationship and the correlation between the two groups.RESULTS：The positive expression rates of Survivin in gastric cancer group and adjacent nontumorous gastric group were 60.4%（29/48） and 22.9%（11/48） respectively （χ2=13.886,P0.05）.Moreover,the expression rate of Survivin in gastric cancer group was higher than that of the adjacent nontumorous gastric group （χ2=13.886,P0.05）.Survivin expression and gastric carcinoma TNM stage were closely related to lymph node metastasis （P0.05）.The positive expression rate of PTTG in gastric cancer group and adjacent nontumorous gastric group was 54.2%（26/48） and 12.5%（6/48） respectively.Moreover,the expression rate of PTTG in gastric cancer group was higher than that of the adjacent nontumorous gastric group （z=-3.890,P0.05）.The positive rate of PTTG was significantly correlated with the differentiation degree,lymph node metastasis and TNM stages （P0.05）.There was a positive correlation between the expression of Survivin and PTTG,as proved by the correlation test （rs=0.555,P0.01）.CONCLUSIONS：The expression of Survivin and PTTG in gastric carcinoma increases in the occurrence and development of gastric cancer play an important role,both the existence of common activation mechanism,constitute a variety of ways to apoptosis of tumor cells.Both can be used as reflected in the biological behavior of gastric cancer and an effective indicator of prognosis.
