摘 要:目的:检测胰腺癌组织中Smad4、p16的表达以及微血管密度(MVD)计数,探讨三者之间的关系。方法:应用免疫组化以及组织芯片技术检测82例胰腺癌组织和8例慢性胰腺炎组织中Smad4、p16和CD34的表达,通过观察CD34来测定MVD。结果:Smad4和p16的阳性率以及MVD在胰腺癌组织中分别为34.1%(28/82)、53.7%(44/82)、19.0±8.9,在慢性胰腺炎组织中分别为100.0%(8/8)、100.0%(8/8)、9.9±4.7;三者在两组间的差异均有统计学意义(P<0.05);Smad4、p16的表达与胰腺癌的分化程度部分有关,MVD与分化程度无关;胰腺癌Smad4和p16的表达均与MVD呈负相关(P<0.05),Smad4与p16的表达呈正相关(P<0.01)。结论:Smad4和p16的表达缺失可能在胰腺癌的发生中起到重要作用,作用方式可能与血管形成相关。OBJECTIVE:To detect the expressions of Smad4 and p16 and the microvasculardensity(MVD) in pancreatic carcinoma tissues so as to understand the relationship among Smad4,p16 and MVD.METHODS:The expressions of Smad4 and p16,CD34(for MVD) were detected by immunohistochemistry and tissue microarray in 82 cases of pancreatic carcinoma and 8 cases of chronic pancreatitis.RESULTS:The positive rates of Smad4,p16 and MVD were 34.1% (28/82),53.7%(44/82) and 19.0±8.9,respectively in pancreatic carcinoma tissues,while they were 100.0% (8/8),100.0%(8/8) and 9.9±4.7 respectively in chronic pancreatitis tissues.The difference was significant between the two groups (P0.05).The expressions of Smad4 and p16 were partly related with the degree of differentiation.In pancreatic carcinoma tissues,the expressions of Smad4 and p16 had obvious negative correlations with MVD (P0.05) and the expression of smad4 had a positive correlation with p16(P0.01).CONCLUSIONS:In pancreatic carcinoma,the loss of Smad4 and p16 may play an important role in pancreatic carcinogenesis.Its underlying mechanisms might be associated with angiogenesis.
