摘 要:目的:研究ZD6474对鼻咽癌细胞株CNE2的抑制作用。方法:采用MTT法测定ZD6474的抑制浓度(IC50),流式细胞仪检测ZD6474的凋亡率及细胞周期分布。结果:MTT数据显示,CNE2细胞的抑制率与ZD6474浓度成正相关,随ZD6474浓度升高,抑制作用越明显,IC50=2.531μmol/L;流式细胞术分析显示,ZD6474处理48h使CNE2细胞凋亡率明显升高(24.02±5.90)%,且随浓度升高凋亡率升高越明显,P<0.001。细胞周期结果显示,ZD6474使细胞周期S期(34.96±5.37)%比例下降,G0/G1期(56.25±1.45)%和G2/M期(17.36±5.73)%比例升高。结论:ZD6474能明显抑制CNE2细胞的增殖,促进其凋亡并改变其细胞周期的分布。OBJECTIVE:To investigate the effect of ZD6474,a tyrosine kinase inhibitor on nasopharyngeal carcinoma CNE2 cell line. METHODS: The antiproliferative and proapoptotie activity of ZD6474 in CNE2 ceil line were detected with MTT method and flow cytometry. RESULTS: With diferent concentrations of ZD6474,a dose-dependent growth inhibition was observed in CNE2 cell line and IC50 was 2.531 μmol/L.Flow cytometry analysis showed that,ZD6474 treatment 48 h to CNE2 cell apoptosis rate increased significantly (24.02±5.90)%,and the apoptosis rate increased with concentration more significantly (P0.001). The results showed that ZD6474 could edcrease the percontage of S phase (34.96±5.37)%,and inereased the percentage of G0/G1 phase (56.25±1.45)% and G2/M phase (17.36±5.73)%. CONCLUSION: This study shows ZD6474 has remarkably anti proliferative and proapoptotie effect on nasopharyngeal carcinoma ceil line CNE2 in vitro.
