脑膜瘤组织MMP-2和MMP-9及组织抑制剂表达临床意义的研究

摘　　要：目的:探讨脑膜瘤组织中基质金属蛋白酶-2、9(MMP-2、MMP-9)及其特异性金属蛋白酶组织抑制剂-1、2(TIMP-1、TIMP-2)基因mRNA的表达及其与肿瘤侵袭和转移性的关系。方法:采用逆转录聚合酶链反应(RT-PCR)法半定量检测36例不同病理类型脑膜瘤组织中MMP-2、MMP-9及TIMP-1、TIMP-2基因mRNA的表达。结果:MMP-2及TIMP-2mRNA在36例脑膜瘤组织中均有表达,除MMP-2在良性组与非典型组间比较差异无统计学意义外(P>0.05),其余各组间比较差异均有统计学意义,P均<0.05。MMP-9及TIMP-1mRNA在36例脑膜瘤组织中也均有表达,各组间比较差异均有统计学意义,P均<0.05。MMP-2/TIMP-2比值在良性组与非典型组间比较差异无统计学意义,P>0.05,而恶性组与其他两组间比较差异均有统计学意义,P均<0.05。MMP-9/TIMP-1比值在3组间比较差异均有统计学意义,P均<0.05。MMPs/TIMPs比值随着肿瘤恶性程度的增加而有逐渐增大的趋势。结论:MMPs/TIMPs基因表达的比值平衡失调在脑膜瘤的侵袭和转移性方面具有重要作用。OBJECTIVE：To investigate the relationship between the expressions of MMP-2,MMP-9,TIMP-1 and TIMP-2 in meningiomas tissues and tumor invision.METHODS：The expressions of MMP-2,MMP-9,TIMP-1 and TIMP-2 mRNA were semiquantitatively detected by reverse transcription-polymerase chain reaction （RT-PCR） in 36 cases of different meningiomas.RESULTS：The positive expressions of MMP-2,MMP-9,TIMP-1 and TIMP-2 were detected in all tumors.The semi-quantitative levels of MMP-2,MMP-9,TIMP-1 and TIMP-2 were significantly different in different groups （P0.05）,but the semi-quantitative level of MMP-2 was not significantly different between the benign group and atypical group（P0.05）.The ratios of MMP-2/TIMP-2 and MMP-9/TIMP-1 were significantly different in different groups （P0.05）,but the ratio of MMP-2/TIMP-2 was not significantly different between the benign group and atypical group （P0.05）.The value of MMPs/TIMPs was gradually increased with the degree increasing of malignant tumors.CONCLUSION：The imbalance of MMPs/TIMPs values shows a significant relevance to brain invasion and metastasis of meningioma.
