摘　　要：目的探讨褪黑素对胃癌小鼠血清caspase-3表达的影响。方法选用6-8周龄SPF级615近交系小鼠,接种小鼠胃癌细胞(MFC),建立荷瘤小鼠模型,并随机分为对照组、褪黑素腹腔注射25mg/(kg·d)组、褪黑素腹腔注射50mg/(kg·d)组、褪黑素腹腔注射100mg/(kg·d)组。对照组小鼠腹腔注射等量生理盐水,药物连续注射1周后取材。应用ELISA检测各实验组小鼠外周血中caspase-3含量的变化。结果胃癌小鼠外周血中caspase-3的含量随褪黑素剂量增大而增加,褪黑素中剂量、大剂量组外周血中caspase-3的含量升高,与对照组和小剂量组相比,有统计学差异。各实验组小鼠肿瘤的体积和质量,随褪黑素剂量增大而减小。结论褪黑素在抑制肿瘤的过程中,具有升高血清caspase-3的作用。褪黑素提高caspase-3的表达,可能是褪黑素抑制肿瘤的作用机制之一。Objective To explore the effect of melatonin on caspase-3 expression in serum of gastric cancer mice. Methods The 6- 8-week-old SPF 615 inbred mice were inoculated with mouse forestomach carcinoma（MFC） ceils to establish the gastric cancer mouse model, then divided into control group, low dose group, middle dose group, and high dose group. The mice were intraperitoneally injected with melatonin 25,50,100mg/（ kg · d） in low, middle, high dose groups, respectively,while the mice were injected with equivalent amount of saline in control group. After continuous injection for a week, the contents of caspase-3 in mouse peripheral blood was detected by ELISA in 4 groups. Results The melatonin increased the caspase-3 contents in a dose-dependent manner. The contents of caspase-3 in middle and high dose groups were significantly higher than that in control group and low dose group. The melatonin decreased the volume and weight of tumors in mice in a dose-dependent manner. Conclusion Melatonin may inhibit the growth of tumor by increasing the serum contents of easpase-3.
