摘 要:目的 分析人肺来源的不同细胞中MAGE-3基因在mRNA水平的表达情况,为肺部良恶性肿瘤的早期鉴别诊断提供研究基础.方法 将人支气管上皮细胞(HBE)、人肺成纤维细胞(HFLF)、原代培养的人肺动脉平滑肌细胞(HPASMC)及人肺腺癌细胞LTEP-a-2同步化培养后,加完全培养基培养24 h后,同步提取各组细胞细胞总RNA,运用实时定量PCR方法分析各细胞中MAGE-3基因在mRNA水平的表达情况.结果 在人肺来源的不同细胞中,正常肺部来源的HBE、HFLF和HPASMC细胞中MAGE-3基因在mRNA的表达水平极低,而人肺腺癌细胞LTEP-a-2中表达丰度明显升高;肺癌细胞中MAGE-3基因在mRNA的表达水平明显高于正常细胞表达水平,差异倍数在20倍以上.结论 肺癌细胞LTEP-a-2中MAGE-3基因mRNA表达水平明显高于肺部正常细胞,可探讨其作为鉴别肺部良恶性病变诊断的恶性病变早期检测指标.Objective To provide research basis for the early differential diagnosis of benign and malignant tumor in lung by analyzing MAGE-3 mRNA expression in different human lung cells. Methods The human bronchial epithelial cells (HBE), human lung fibroblasts (HFLF), primary cultured human pulmonary artery smooth muscle cells (HPASMC) and human lung adenocarcinoma cells (LTEP-a-2)were synchronized cultured with complete medium for 24 hours,the total cellular RNA was simultaneously extracted,and MAGE-3 mRNA expression was analyzed by real-time polymerase chain reaction. Results MAGE-3 mRNA expression was extremely low in HBE, HFLF and HPASMC,and significantly increased in LTEP-a-2. MAGE-3 mRNA expression in lung cancer cells was significantly higher than that in normal cells. Conclusions MAGE-3 mRNA expression in LTEP-a-2 is significantly higher than that in normal lung cells,which could be an early detection indicator for differential diagnosis of benign and malignant lung lesions.
