摘 要:目的探讨mtDNA(mitochondrial DNA,mtDNA)np16189和np16223多态性及其单倍型与乳腺癌的相关性。方法采用聚合酶链反应-限制性片段长度多态性(polymerase chain reaction-restricted fragment lengthpolymorphism,PCR-RFLP)技术对195例乳腺癌患者和196例正常对照的mtDNA np16189和np16223基因型进行检测,计算基因型频率,应用SHEsis软件构建单倍型,利用χ2和OR(Odds ratio)检验分析各位点及其单倍型与乳腺癌的相关性。结果 mtDNA np16189和np16223基因型频率及两者构建的单倍型频率在两组中的分布差异均无统计学意义(P>0.05)。结论 mtDNA np16189和np16223多态性可能与乳腺癌无相关性。Objective To explore the association between mtDNA np16189,np16223 polymorphisms and breast cancer.Methods 195 sporadic patients with breast cancer and 196 healthy controls were recruited from the first affiliated hospital of Ningxia medical university from Nov.2008 to Sep.2009.The average age of two groups were 48.58±10.08 and 47.16±9.68 years respectively.The PCR-based RFLP assays were conducted to determine the genotype of np16189 and np16223.The Odds ratio(OR) and 95% confidence interval(CI) were used to estimate the association between risk and breast cancer.SHEsis software was used to analyzed the haplotypes that composed by two sites.Results ①There were no significant difference of np16189 and np16223 genotype distribution between cases and controls(P0.05).②There were no significant difference of haplotypes distribution between cases and controls(P0.05).Conclusion mtDNA np16189 and np16223 polymorphysims and their haplotypes may be not associated with breast cancer in Ningxia Han population.
