中药复方松友饮抑制姑息性肝切除术后残癌生长和转移的实验研究

摘　　要：目的:研究中药复方松友饮对姑息性肝切除裸鼠模型中残癌生长及转移的影响。方法:以人肝癌细胞(MHCC97H)建立裸鼠移植瘤姑息性肝切除模型,36只裸鼠随机分成对照组和松友饮组,每天1次灌胃给药连续5周,每周测体质量。给药结束后第2d每组处死6只裸鼠观察肿瘤大小、肺转移等;并检测残癌中白细胞介素-1β(IL-1β)和增殖细胞核抗原(PCNA)表达情况。剩余裸鼠用于观察生存期。结果:松友饮组与对照组残癌体积为(724.9±337.6)mm3和(1205.2±581.3)mm3(P<0.05),抑瘤率为39.9%;肺转移分级下降(P<0.05)。前者体质量大于对照组(P<0.001),生存期延长率为22.7%;IL-1βmRNA表达水平显著下调(P<0.001),蛋白水平为(18.0±5.1)pg/mL和(27.9±4.5)pg/mL(P<0.01);PCNA指数为(75.2±8.5)%和(86.1±4.8)%(P<0.001)。结论:松友饮灌胃给药下调残癌表达IL-1β和PCNA,抑制残癌生长、转移,对肝癌病人有潜在的应用价值。Objective： To investigate the effects of Chinese herbal compound ＂Songyou Yin＂ on tumor growth and metastasis of residual hepatocellular carcinoma （HCC） following palliative liver resection in nude mice model. Methods： HCC models of palliative resection were established by orthotopic implantation of human HCC cell line MHCC97H xenografts. Thirty - six nude mice bearing residual HCC were randomized into control group and ＂Songyou Yin＂ group. Daily intragastric adminstration of ＂Songyou Yin＂ was given for 5 consecutive weeks 24 hours after palliative liver resection. Body weight was recorded weekly. Six mice in each group were sacrificed on the second day after treatment. Tumor volume, lung metastatic extent （grade） , and the expression of IL-1β and proliferating cell nuclear antigen （PCNA） were examined. The remaining mice were used to observe life span. SAS 8.2 software was used for statistical analysis, and the significance was defined as P〈0.05. Results： The residual HCC tumor volume was（724.9±337.6）mm3 and（1205.2±581.3）mm3 in ＂Songyou Yin＂ group and control group respectively （P〈0.05） , with tumor inhibition rate of 39.9%; the lung metastatic extent （grade） reduced significantly in ＂Songyou Yin＂ group （P〈0.05）, body weight of mice in ＂Songyou Yin＂ group was heavier than that in control group （P〈0.001）. IL-1β mRNA relative expression was significantly downregulated as comparison was made between two groups （P〈0.001）, IL- 1β protein level （Pg/mL） was（18.0±5.1）and（27.9±4.5）（P〈0.01）, and the PCNA index （%） was（75.2±8.5）and（86.1±4.8）（P〈0.001） respectively at 5th week. The life span of mice in ＂Songyou Yin＂ group was significantly longer than that in the control, being（62.1±2.0）and（76.2±3.2d）（P〈0.05）, the rate of prolong-life was 22.7%. Conclusion： Five weeks administration of Chinese herb ＂Songyou Yin＂ resulted in inhibition of tumor growth, delayed body weight loss
