摘 要:目的:探讨鼻咽癌细胞株(CNE1、CNE2)间不同辐射敏感性与其G2期阻滞的关系。方法:采用流式细胞仪检测不同剂量X线及照射后不同时间点CNE1、CNE2细胞G2期细胞比例的变化。结果:2Gy或5GyX线照射后,CNE1,辐射相对抗拒株,在1、4、8、24h时间点,检测到明显G2期阻滞,且有时间剂量效应(P<0.05);CNE2,辐射相对敏感株,照射后24h内G2期细胞比例升高不明显,与CNE1有显著意义(P<0.05),CNE2在48h点才开始出现G2期阻滞。同时,CNE1、CNE2均未见明显细胞凋亡(P>0.05)。结论:电离辐射后发生的G2期阻滞的不同可能是鼻咽癌细胞株(CNE1、CNE2)间内在的辐射敏感性的差异原因之一。Objective To explore the relationship between different inherent radiosensitivity and variances of G2 arrest in NPC cell lines (CNE1 and CNE2). Methods Flow cytometry was used to detect the changes of G2 phase in CNE 1 and CNE2 cell cycle at different time points as the two cell lines were exposed to different doses of X-ray irradiation. Results In CNE1, which was more radioresistant to irradiation than CNE2, 2 Gy or 5 Gy X-ray irradiation resulted in G2 arrest in a time- and dose-dependent manner (P 〈 0.05) at hours 1, 4, 8, and 24 after irradiation (P 〈 0.05); while in CNE2, there was no obvious G2 arrest at hours 24 (P 〈 0.05). CNE2 cells at G2 phase became accumulated at hour 48. No evident radiation-induced apoptosis was detected in both CNE1 and CNE2 (P 〉 0.05). Conclusions The various levels of ionizing radiation to induce G2 arrest may be one of the causes for different inherent radiosensitivity between NPC cell lines CNE1 and CNE2.
