摘 要:目的:探索中国汉族人群(Klotho,KL)基因G-395A、F352V及C370S单核苷酸多态性(Single nucleotide polymorphisms,SNP)及其与心动周期信号谱特征的相关关系。方法:随机选择汉族自然人群个体177为研究对象[男96,女81例,平均年龄(65.11±10.7)岁],获取其静脉血白细胞基因组DNA。采用等位基因特异性引物PCR技术检测Klotho基因3个位点的基因型,利用《HPS谱及其混沌特征分析系统软件》检测人群的卧位心动周期信号谱(Heart period signal,HPS)参数。结果:①KL基因G-395A SNP AA基因型携带者其绝对高频功率(Absolute high frequency power,AHP)及相对高频率(Relative high frequency power,RHP)参数较其GA携带者降低,均有显著性差异(P<0.05);同时携带AA基因型者的RHP参数也较GG携带者降低,亦有显著性差异(P<0.05)。②KL基因F352VSNP VV突变基因型携带者其相对超低频率(Relative ultra-lower frequencypower,RUP)、超低频/高频比值(RatioofAUP toAHP,RUH)参数值较FF野生型纯合子基因型携带者低,差异有显著性(P<0.05);携带VV基因型者绝对低频功率(Absolute lowfrequency power,ALP)、AHP、RHP以及总功率(Total power value,TPV)较其FF野生型携带者高,也有显著性差异(P<0.05);汉族人群KL基因F352V多态性的突变型VV基因型纯合子携带者其RUP、RUH参数值也较其FV杂合子携带者降低;突变型VV基因型纯合子携带者其RHP参数值则较FV杂合子携带者高,亦有显著性差异(P<0.05)。③KL基因C370S多态性的突变型SS基因型纯合子携带者其低频/高频比值(Ratio of absolute low frequency power to AHP,RLH)参数值较CC野生型纯合子携带者低有显著性差异,同时也较CS杂合子携带者低,其差异亦有显著性(P<0.05)。结论:KL基因G-395ASNP突变型AA与AHP、RHP减低有关;F352VSNP突变型VV与RUP、RUH减低有关,亦与其ALP、AHP、RHP以及总功率(Totalpower value,TPV)增高有关,这就提示klotho基因多态性与心动周期信号谱特征的有明确相Objective:To search for association of heart period signal spectrum features in the supine posture with single nucleotide polymorphisms of three sites of klotho G-395A,F352V and C370S in Chinese Han nationality population. Method s:Genomic DNA of white blood cells from 177 individuals as research objects ( female 81 and male 96,age 65.11±10.7 y)selected randomly from Chinese Han nationality population was gotten. The SNPs of klotho G-395A,F352V and C370S were genotyped by allele-specific primer(ASP) PCR technique. We applied the analytical software for spectral features of heart period signal (HPS)to determine the spectral parameters of heart period signal in the supine posture in this group. Results:①The spectral parameters of HPS including absolute high frequency power(AHP)and relative high frequency powe(rRHP)in the group with AA genotype were significantly low than those in the group with GA genetype of klotho G-395A ( P0.05). The spectral parameter of relative high frequency power(RHP)in the group with AA genotype were significantly lower than those in the group with GG genetype of klotho G-395A(P0.05). ②The spectral parameters of HPS including relative ultra-lower frequency power(RUP)and ratio of AUP to AHP(RUH)in the group with VV genotype were significantly lower than those in the group with FF genetype of klotho F352V(P0.05). The spectral parameters of absolute low frequency powe(rALP),absolute high frequency power(AHP),relative high frequency power(RHP)and total power value(TPV)in the group carrying VV genetype were significantly higher than those in the group with FF genetype of klotho F352V(P0.05). The spectral parameters of RUP and RUH in the group with VV genotype were obviously lower than those in the group with FV genetype of klotho F352V and the value of RHP in the group with VV genotype were also much higher than that in the group with FV genetype of klotho F352V(P0.05). ③The value of ratio of ALP to AHP (RLH)in
