摘  要:目的应用银杏叶提取物阻断大鼠胃黏膜癌前期病变的发展并观察胃黏膜原癌基因表达的变化。方法将80只4周龄Wistar大鼠随机分为4组:正常组、模型组、银杏叶提取物低、高剂量组,每组20只,以喂饲100μg/mLMNNG溶液的方法制备大鼠胃黏膜癌前期病变的动物模型。在制备大鼠胃黏膜癌前期病变模型过程中采用不同浓度的银杏叶提取物对治疗组进行干预(低剂量组0.5mg·Kg-1·d-1、高剂量组1.5mg·Kg-1·d-1),共20周。第30周末,处死所有大鼠,观察胃黏膜组织病理学变化、癌基因(bcl-2、c-myc等)的表达。结果与模型组比较,银杏叶提取物低、高剂量组肠化生、异型增生发生率低,差异有统计学意义(P<0.01)。随着病变程度不同,各癌基因的表达不同。模型组与正常组及干预组比较,bcl-2、c-myc、FasL的表达明显增加,而Fas的表达减少(P<0.01或0.05)。结论银杏叶提取物能够调节细胞的增殖和凋亡,有效地阻断胃黏膜癌前期病变的发生、发展。Objective To investigate the effect of Ginkgo biloba extract on gastric precancerous lesions and the change of expression of oncogenes ( bcl-2,cmyc,Fas,FasL) in gastric precancerous lesions in Wistar rats. Methods Eighty Wistar rats ( 4 weeks old) were randomly divided into four groups: normal group,model group,low and high dose groups. MNNG ( 100 μg/mL) was used feeding rats to make animal models of gastric precancerous lesions ( 20 weeks) ,and low/high dose groups were lavaged with Ginkgo biloba extract in 0.5 mg·Kg -1·d -1 and 1.5 mg· Kg -1·d -1,respectively. After 30 weeks,all rats were killed. Histopathological changes of gastric mucosa in these rats were observed. Expression of oncogenes were also determined. Results The incidence of mild to severe metaplasia, dysplasia in low and high dose groups were significantly lower than those in model group ( P 0. 01) . Compared with normal group,low and high dose groups,the expression of bcl-2,c-myc,FasL increased in model group,but the expression of Fas decreased. There were statistic differences among different groups ( P 0. 01 or 0. 05) . Conclusion Ginkgo biloba extract can regulate cell proliferation and apoptosis,effectively intervene in the development of gastric precancerous lesions in rats.
