摘  要:目的:研究转染喉鳞状细胞癌总RNA对树突状细胞(DC)免疫功能的影响,观察肿瘤总RNA修饰后的DC在体外诱导高效而特异的抗喉癌免疫效应。方法:采用分离脐血单核细胞体外诱导DC,Trizol法提取喉鳞状细胞癌细胞株Hep-2总RNA后转染入DC,流式细胞仪分析DC表面分子CD40、CD80、CD83、CD86的表达,LDH法评估转染肿瘤总RNA的DC瘤苗的细胞毒性T淋巴细胞反应。结果:转染后的DC表面分子表达CD40(60.6±0.15)%,CD80(60.2±0.21)%,CD83(85.1±0.06)%,和CD86(91.9%±0.09)%,而在未转染的未成熟DC中低表达;转染RNA的DC瘤苗对于Hep-2细胞株有特异性杀伤效应,在E/T为40:1时,杀伤效率达45.6%,而对对照组靶细胞杀伤效率较低。结论:喉鳞状细胞癌总RNA转染修饰的DC能特异性诱导细胞毒性T淋巴细胞,显著提高DC的抗原提呈功能,在体外能诱导高效而特异的抗喉癌免疫效应。Objective:To investigate the effect of dendritic cells(DCs) transfected with laryngeal squamous cell carcinoma cell line Hep-2 cells total RNA,and to observe the specific anti-laryngeal cancer immunological results induced by modified DC in vitro.Methods:DCs were generated from the cord blood monocytes.Total RNA was isolated from laryngeal squamous cell carcinoma cell line Hep-2 cells by Trizol,and was transfected into DCs.The expression of cell surface molecules CD40,CD80,CD83,and CD86 were detected using flow cytometry.The competence of inducing specific cytotoxic T lymphocyte(CTLs) was assessed with LDH.Results:DC surface molecules CD40(60.6 ±0.15) %,CD80(60.2 ± 0.21) %,CD83(85.1 ± 0.06) %,and CD86(91.9% ± 0.09) % were highly expressed in Hep-2 RNA-DC than that in untreated DC.The cytolytic rate to laryngeal cancer cell was 45.6% at 40:1 E/T ratio.However,no significant cytolytic activity was seen in control cell lines.Conclusion:DCs transfected with Hep-2 cells total RNA could induce specific cytotoxic T lymphocytes,increase the antigen presenting function of DC,and induce specific CTL killing activity to laryngeal squamous cell carcinoma.
