摘 要:目的:利用量子点(QDs)免疫荧光双标法检测人不同宫颈病变组织中窖蛋白(Cav-1)和增殖细胞核抗原(PCNA)蛋白的共表达,并探讨Cav-1蛋白与宫颈鳞癌临床病理参数的关系。方法:利用QDs免疫荧光组织化学技术检测人不同宫颈病变组织[正常宫颈上皮10例,宫颈上皮内瘤变(CIN)Ⅰ10例,CINⅡ-Ⅲ10例,宫颈鳞癌70例]中Cav-1蛋白的表达,并利用QDs免疫荧光双标法同时检测Cav-1和PCNA蛋白的共表达。结果:Cav-1蛋白在正常宫颈鳞状上皮、CINⅠ、CINⅡ-Ⅲ和宫颈鳞癌中阳性率分别为:0,20.00%,40.00%和55.71%,正常宫颈鳞状上皮和CINⅠ分别与宫颈鳞癌相比,Cav-1蛋白表达的差异均有显著性(P<0.05),而其他组别之间比较,差异均无显著性(P>0.05)。Cav-1蛋白表达与宫颈鳞癌的临床病理参数均无关(P>0.05)。但在宫颈上皮内瘤变和宫颈鳞癌组织中Cav-1和PCNA蛋白表达之间呈显著正相关(P<0.01,r=0.429)。结论:Cav-1蛋白表达有助于细胞增殖,与宫颈病变的恶性进展密切相关。Objective:To detect the co-expression of caveolin-1(Cav-1)and PCNA proteins by aquantum dots(QDs)double-labeling immunofluorescence method in human different cervical lesions,and to investigate the relationship between expression of Cav-1protein and clinicopathologic parameters of cervical squamous cell carcinomas.Methods:QDs immunofluorescence histochemistry technology was used to detect the expression of Cav-1protein in human different cervical lesions including 10cases of normal cervical epithelium,10cases of low-grade cervical intraepithelial neoplasia(CINⅠ),10cases of high-grade cervical intraepithelial neoplasia(CINⅡ-Ⅲ)and 70cervical squamous cell carcinomas,and co-expression of Cav-1and PCNA proteins was simultaneously detected by a double-labeling immunofluorescence.Results:The positive rate of Cav-1protein in the normal cervical epithelium,CINⅠ,CINⅡ-Ⅲand cervical squamous cell carcinomas was 0,20.00%,40.00%,and 55.71%,respectively,and the differences were significant between normal/CINⅠcervical epithelium and cervical squamous carcinoma(both P0.05).The positive rate of Cav-1protein was not related with clinicopathologic parameters of cervical squamous cell carcinoma(P0.05).But a positive correlation was observed between Cav-1and PCNA protein expression in the CIN and cervical squamous cell carcinomas(P0.01,r=0.429).Conclusion:The expression of Cav-1protein may contribute to cell proliferation,and correlate closely with the malignant progression of cervical lesions.
